To understand the molecular functions and gene networks of Auxin Response Factor (ARF) and Lateral Organ Boundaries Domain (LBD) transcription factors for regulating lateral root (LR) development in plant root architecture, we investigated molecular mechanisms of LBD18 action during LR initiation, primordium development, and emergence, a positive feedback loop of auxin-responsive ARF7/ARF19-LBD18, the role of ARF7/ARF19-LBD16/LBD18 gene network during adventitious root formation, and the roles of other LBD genes in LR development, using Arabidopsis as a model plant.
□ Results
1. Elucidation of LBD18 functions in LR initiation, primordium, and emergence.
1-1. Functional analysis duing LR initiation and primordium development.
1-2. Expression analysis of cell cycle genes.
1-3. In planta functional analysis of dimerization domain of LBD16 and LBD18.
1-4. Geneome-wide analysis of target genes regulated downstream of LBD18.
2. Analysis of gene regualtory networks of auxin-LAX3-ARF7/ARF19-LBD18.
2-1. Characterization of double positve feedback loops of LBD18 and ARF19.
2-2. Analysis of a positive feedback loop mediated by protein-protein
intearctions between LBD18 and ARFs.
3. Role of ARF7/ARF19-LBD16/LBD18 gene network in adventious root formation.
4. Characterization of the roles of LBD13 and LBD14 during LR formation.
5. Characterization of the roles of LBD37, LBD38, and LBD39 during LR formation.
□ Expected Contribution
The present study reveals the roles and mechanisms of LBD transcription factors in gene regulatory networks of LR development duing the auxin response and the double positive feedback loops. The roles of other LBD genes in lateral root formation and the signlaing pathways and molecular mechanisms involved in LR development in response to cold are also identified. Nevertheless, genome-wide identification and functional analysis of direct tagets of LBD18 need to be followed. Other plant hormones, such as brassinosteroids, and signaling peptide hormones, app2ear to interconnect with auxin signaling pathway during LR development. Crosstalk among these plant homones during LR development need to be investigated to get better insights into how to improve plant root archtecture for enhance crop yield.
