연구의 목적 및 내용
1세부과제에서는 치료용 단백질 생산 형질전환 소 개발을 위하여 본 과제의 목적은 RNA-Guided ENdonuclease(RGEN) 시스템 미세주입에 의한 치료용 단백질 생산용 Knock-in 벡터를 개발하고 그 벡터의 검증 및 knock-in 수정란과 미세주입에 의하여 생산된 산자를 분석하는 데 있다. 1협동과제에서는 사람 섬유아세포성장인자2(hFGF2)가 넉인된 형질전환 소 개발을 위해 1세부에서 개발한 RGEN용 hFGF2 knock-in 벡터를 이용하여 유전자주입 수정란 생산 시스템을 구축하고 형
연구의 목적 및 내용
1세부과제에서는 치료용 단백질 생산 형질전환 소 개발을 위하여 본 과제의 목적은 RNA-Guided ENdonuclease(RGEN) 시스템 미세주입에 의한 치료용 단백질 생산용 Knock-in 벡터를 개발하고 그 벡터의 검증 및 knock-in 수정란과 미세주입에 의하여 생산된 산자를 분석하는 데 있다. 1협동과제에서는 사람 섬유아세포성장인자2(hFGF2)가 넉인된 형질전환 소 개발을 위해 1세부에서 개발한 RGEN용 hFGF2 knock-in 벡터를 이용하여 유전자주입 수정란 생산 시스템을 구축하고 형질전환수정란 생산 배양기술을 개발하며 hFGF2 knock-in 유전자 주입된 수정란 생산 효율 제고에 있다. 2협동과제의 목적은 사람 사이토카인 생산용 형질전환 동물을 생산하기 위한 knock-in 수정란 생산하는데 있으며 이를 위하여 RNA-Guided ENdonuclease(RGEN)용 사람 사이토카인 생산용 knock-in 벡터를 젖소 수정란에 미세주입하는 기술을 개발하였으며, knock-in 수정란 검증 및 knock-in 수정란을 이식을 실시하는데 있다. 3협동과제에서는 소 전핵기 수정란 내 엔도스타틴 또는 락토페린 knock-in 벡터 및 RGEN의 미세주입에 의한 착상전 배발달 및 유전자 적중 여부 확인, 미세주입에 의해 생산된 소 배반포의 동결보존 및 이식, 미세주입 된 소 배반포의 이식을 통한 산자 생산 및 형질전환 젖소의 확보를 목적으로 하고 있다.
연구개발성과
1세부과제에서는 젖소 β-casein 엑손 2을 target으로 하는 RGEN용 락토페린, FGF2와 사이토카인 knock-in 벡터 4종을 개발하였다. 젖소 β-casein 엑손 7을 target으로 하는 RGEN용 엔토스타틴 knock-in 벡터 3종을 개발하였다. 수정란 내 RGEN 주입에 의한 상동유전자재조합 검증용 벡터 16종을 개발하였다. 젖소 β-casein 엑손 2 및 7을 target 하는 RGEN을 확보하고 in vitro assay, in vivo assay에서 정상적으로 작동함을 확인하였으며 수정란에서 50%의 indel 을 확인하였다. 유선세포에서 knock-in 벡터의 발현을 RT-PCR에 의하여 검증하였다. 치료용 단백질 유전자를 one step으로 삽입할 수 있는 knock-in 벡터 개발2종 이상 개발하였다. knock-in 수정란 검증을 위한 PCR control 벡터를 구축하고 PCR 조건 확립하고 검증하였다. DT 선별 marker가 없는 knock-in 벡터의 5종 이상을 구축하였다. knock-in 수정란 검증하여 hFGF 유전자에 대해서는 약 4.8%, hIL11유전자에 대해서는 약 19%, endostatin 유전자에 대해서는 평균 10.8%, lactoferrin 유전자에 대해서는 평균 18%의 넉-인을 확인하였으며 넉-인된 염기서열을 확인한 결과 모두 정상적으로 넉-인이 되어 있었다. knock-in 유선세포로부터 치료용 단백질의 활성 검증을 실하였다. 현재 knock-in 벡터을 주입한 수정란으로부터 태어난 산자 2마리와 말기 유산된 1마리로부터 넉-인 여부를 검증하였다.
1협동과제에서는 형질전환 수정란 생산시스템 구축을 위해 간편하면서도 효율적인 Triladyl과 Sperm filter를 이용한 체외수정 방법을 개발하였다. 형질전환 수정란 발달 제고를 위해 난자수와 배양 드롭 크기의 영향을 조사하여 5 ~10 개의 난자 배양 시 10 ul 드롭이 배발달에 효과적임을 확인하였고 적정량의 성장인자(IGF+EGF) 와 항산화제(Flavonoid)의 첨가 효과를 확인하였으며, 0.1 uM Allicin 또는 1 uM β-cryptoxanthin을 체외성숙용 배양액에 첨가하여 체외 발달을 증진시킬 수 있음을 확인하였다. 또한 Time-lapse 모니터링 장비로 우수 수정란 선별 시스템을 새롭게 구축하였다. hFGF2 knock-in 유전자가 주입된 형질전환 수정란 생산은 pBSK(-)_RG1_bBCE3_hFGF_GFP KI vector I (200)/ bBCE3 sgRNA (100)/ pBSK(-)#1 sgRNA (100)/ Cas9 mRNA (400)/ bRAD51 mRNA (100)/ Nuclease free water with Scr7 (1000)을 혼합하여 주입한 후 24시간 동안 RS-1 처리하는 것이 효율은 높지 않지만 hFGF2 넉인된 형질전환 수정란 생산이 가능한 조건임을 최종적으로 확인하였다.
제2협동과제에서는 젖소 체외수정란 전핵에 knock-in vector의 미세주입 방법 개발과 체외수정란의 배반포로의 발달율 증진을 이루어 가능한 형질전환 배반포를 생산하였다. 또한, 젖소 체외수정란 세포질에 knock-in vector를 미세주입하는 방법의 개발과 이를 이용한 형질전환 수정란 생산 및 knock-in 수정란 유래 배반포의 동결보존 방법 개발하였다. 이들 기술을 바탕으로 젖소 체외수정란 세포질에 two sgRNA RG1 bBCE3 HIL11 GFP (-DT)K1 Vector I을 주입하였고, 발달된 배반포중에서 GFP의 발현과 PCR 검증을 통하여 약 30%에서 성공적으로 Knock-in이 확인 되었다. 생산된 형질전환 수정란은 대리모에 이식하여 성공적인 임신을 확인하였기에 Knock-in 송아지 생산을 기대할 수 있다.
제3협동과제에서는 엔도스타틴 또는 락토페린 유전자 Knock-in 벡터 및 RGEN의 미세주입, 미세주입 소 수정란의 체외배양 및 생산, 미세주입 배반포에서 유전자 적중 여부 분석, 미세주입 소 배반포의 동결 및 융해, 미세주입 소 배반포의 이식, 임신진단 및 생산을 실시하였다.
연구개발성과의 활용계획(기대효과)
본 연구에서 개발된 knock-in 벡터와 유전자 가위를 활용한 시스템은 젖소 β-casein 유전자 위치에서 다양한 치료용 단백질을 생산하는 데 활용될 수 있다. 특히 본 과제에서 개발된 one-step 벡터를 이용하면 목적 유전자를 cloning 한 후 sub-cloning 방법에 의하여 바로 knock-in 벡터에 삽입할 수 있어서 시간과 경비를 절약하면서 목적으로 하는 knock-in 벡터를 매우 수월하게 제작할 수 있다. 또한 본 과제에서 획득한 유전자 가위의 합성과 활용에 대한 노하우는 다양한 가축의 유전자 편집에 활용이 가능하다. 본 과제에서 개발된 방법을 이용하면 hFGF2 넉인된 형질전환 수정란 생산이 가능하고 개발된 체외배양 시스템과 우수 수정란 선별시스템을 활용하면 효율적인 형질전환동물 생산이 가능하다. 또한 본 과제에서 구축한 소 수정란의 knock-in 기술은 각종 형질전환 산자생산 뿐만 아니라 외래 유전자의 전이 과정이 요구되는 각종 연구 등에 활용할 것이며, 개발된 knock-in 형질전환기술은 형질전환 동물의 생산에 있어서 큰 문제점으로 대두되고 있는 외래 유전자의 지속적인 발현에 의한 생리적인 부작용을 최소화할 수 있는 수단으로서 차후 생산하고자 하는 생리활성물질을 발현하는 형질전환 동물개발 연구에 지속적으로 활용이 가능할 것이다

The purpose of sub-project 1 is to develop a knock-in vector for the production of therapeutic protein by microinjection of RNA-Guided ENdonuclease (RGEN) system and to analyze knock-in embryo and cattle produced by the microinjection of knock-in vector and RGEN. The purpose of cooperative project 1 is to develop a transgenic cow for human fibroblast growth factor 2 (hFGF2) created from transgenic embryos using the RGEN hFGF2 knock-in vector, culturing technology and enhancing production efficiency of hFGF2 knock-in gene injected embryos. The purpose of cooperative project 2 is to produce and transfer knock-in mediated RNA-Guided ENdonuclease(RGEN) bovine embryos for human cytokine production. RNA-Guided ENdonuclease(RGEN) knock-in vector was micro-injected into embryos of dairy cattle and knock-in efficiency is evaluated. The purpose of cooperative project 3 is to identify of preimplantation embryo developmental compentence and genes targeting by microinjection of endostatin and lactoferrin knock-in vector and RGEN into zygotes, and to carry out cryopreservation and transfer of microinjected blastocysts and production of transgenic dairy cattle by microinjected blastocysts.
Results
We developed four kinds of knock-in vector for human lactoferrin, hFGF2 and cytokine to target cattle β-casein exon 2 using RGEN.
Three kinds of endostatin knock-in vector to target cattle β-casein exon 7 developed. The sixteen kinds of knock-in vector for verification of homologous recombination by RGEN injection into zygotes were developed. We developed RGEN targeting cattle β-casein exon 2 and 7 and validated RGEN activity in an in vitro and in vivo assay. The indel level of 50% was observed in the embryo injected with RGEN. Expression of knock-in vector in mammary gland cells was confirmed by RT-PCR. We have developed two or more knock-in vectors that can insert the therapeutic protein gene in one step. PCR control vectors for each knock-in vector were constructed and PCR condition for screening of knock-in were established. We constructed more than five kinds of knock-in vectors without DT marker. The efficiency of knock-in embryos was 4.8% for hFGF knock-in vector, about 19% for hIL11 knock-in vector, 10.8% for endostatin knock-in vector and 18% for lactoferrin knock-in vector. As a results of sequence analysis, the embryo was conformed knock-in embryo. We examined the activity of therapeutic proteins from knock-in mammary gland cells. We analyzed two live calves and one aborted fetus by PCR to determine whether or not knock-in.
Simple and efficient in vitro fertilization method using Triladyl and Sperm filter was developed to construct the transgenic embryo production system. In order to improve the development of transgenic embryos, we investigated the effect of the different culture drop size on the growth of 5 or 10 oocytes, and 10 ㎕ drop was effective for embryo development. The optimized concentration of growth factors (IGF + EGF) and antioxidant Flavonoid was effective for embryo development. Also, 0.1 μM Allicin or 1 μM β-cryptoxanthin could be added to the culture medium for in vitro maturation to enhance the in vitro embryo development. In addition, we have newly constructed a screening system to select normal and healthy transgenic embryos with time-lapse monitoring equipment.
The production of transgenic embryos was possible through the optimized protocol in which the hFGF2 knock-in gene was microinjected in embryos, mixed using pBSK (-) RG1_bBCE3_hFGF_GFP KI vector I (200) / bBCE3 sgRNA (100) / pBSK (-) # 1 sgRNA (100) / Cas9 mRNA (100) / Nuclease free water with Scr7 (1000), and then cultured in RS-1 for 24 hours.
Embryo transfer available blastocysts were produced by using micro-injection of knock-in vector into pronuclear of embryos of dairy cow. Additionally, micro-injection of knock-in vector into cytoplasm of embryo and cryopreservation methods were developed. based on developed methods, two sgRNA RG1 bBCE3 HIL11 GFP (-DT)K1 Vector I was injected into cytoplasm of embryos and 30% of developed blastocysts were successfully knock-in by evaluating expression of GFP and PCR analysis. Therefore, further process such as transfer of knock-in embryos and production of transgenic cattle will be available. After microinjection of endostatin or lactoferrin knock-in vector, the developmental rate to blastocyst was about 20%. In order to preserve microinjected blastocysts through ultracold cryopreservation and to improve low survival rate, we confirmed the improvement of quality using melatonin. We conducted Transfer of microinjected blastocysts, pregnancy diagnosis and production of calf.
Expected Contribution
The knock-in vector and RGEN system developed in this study can be used to produce various therapeutic proteins at the β-casein gene position in the cattle. In particular, the one-step vector developed in this project can clone the target gene and insert it into the knock-in vector directly by sub-cloning method. It can be produced easily. The know-how on the synthesis and utilization of RGEN obtained in this project can be used for gene-editing of various livestock. Using the method developed in this project, it is possible to produce hFGF2 knock-in transgenic embryos and it is possible to produce transgenic animals efficiently by using the developed in vitro culture system and the screening system of excellent embryos. Established transgenic embryo production technique will be useful for producing other transgenic animal and investigation of transgenesis mechanism. Our developed technique in this research will reduce physiological disadvantages on transgenic animal derived from continues expression of inserted genes. Thus, the results of in this study will offer stable and safe methods for producing transgenic animal production.
