□ Results
- We found that the high concentration of mulberry leaves’ and fruit’s extract had an anti-obesity effect, and the degree of anti-obesity effect was also different according to variety.
Anthocyanin rich fraction (ARF) was fractionated and purified from fruits extract by open chromatography using PB-600 macroporous resin. FRF showed 35.46 mg/g of total flavonoid content, 32.16 mg/g of rutin. Total anthocyanin contents was 44.36 mg/g in the mulberry extracts, whereas anthocyanin content was increased to 185.60 mg/g in ARF. FRF and ARF were used in a bioassay to evaluate the cell toxicity using Raw 264.7 mouse macrophage cell viability test and measured for anti-obesity effect and its related gene expression in lipid metabolism using real-time PCR and western blot analysis.
The experiments were performed at a concentration of less than 1000 ㎍/㎖, which is not cytotoxic by MTT assay with 3T3-L1 preadipocyte. In Oil Red O staining, FRF and ARF (1000 ㎍/㎖) treatments showed 35.39%, 46.44% of inhibition rate in lipid accumulation compared to the control respectively. The triglyceride content of 3T3-L1 adipocyte treated by FRF and ARF (1000 ㎍/㎖) were reduced to 23.22% and 59.32%, and free glycerol content was increased to 106.04% and 107.05% by AdipoRedTM assay. FRF containing rutin and ARF containing anthocyanin affected gene expresstion of mRNA. FRF and ARF were found to have decrease the gene expression of SREBP-1c, PPAR, C/EBP α, FAS, ACC in a dose-dependent manner. Rutin was contributed as major constituent in FRF for inhibition of SREBP-1c gene expression, also it was affected that sub-factor of SREBP-1c, PPARγ, C/EBP/α, FAS and ACC were inhibited. ARF comprising to both C3G with C3R was reduce to a higher level than C3G alone in the gene expression of lipid metabolism related gene. Western blot analysis confirmed that lipid metabolism related gene was markedly decreased.
Anthocyanins and flavonoids from mulberry (Morus alba L.) prevent the development of obesity in mice fed a high-fat diet(HFD). In this study, our purpose was to explore the effect of the administration of anthocyanin rich (MPA) and flavonoid rich fraction (MFRF) on the development of obesity in mice fed a high fat diet (HFD).
Four-week-old C57BL/6 mice were fed a high-fat diet (HFD) with or without MPA and MFRF for 12 weeks. Body weight, serum and hepatic lipids, liver and adipose tissues morphology, insulin and leptin were assessed. MPA and MFRF suppressed the body weight gain of the HFD-fed C57BL/6 mice. Intake of MPA and MFRF reduced body weight for the HFD-fed mice. Furthermore, MPA and MFRF could significantly decrease TG in the liver, and inhibit leptin secretion. MPA and MFRF could also improve insulin resistance. Moreover, MPA and MFRF exerted potentially anti-inflammatory effect. Therefore, MPA and MFRF could be used to help counteract obesity.
Anthocyanin was spray-dried after mixing with WPI,anthocyanin capsules were prepared in powder form. The particle size of the AWM is 100 to 300 ㎛, showed the appearance of a spherical shape. Ratio of anthocyanins present in the capsule was about 83%.
Whereby the encapsulation efficiency could not be eluted with a solvent seems to test. When compared with the release control is 0 hours, the amount of anthocyanins was increased after 5 hours. It can be seen that the AWM and control in the release of the anthocyanin. Thus, encapsulation of anthocyanins was protected from environmental factors, such as thermal degradation that can occur in the thermal process,structural change due to the pH change and energy transfer,which occurs during the light exposure. It was protected from destruction and discoloration by the additive, such as sugars, organic acids and metal ions. When used as raw material for foods or cosmetics to AWM conditions can be established to minimize the destruction of the anthocyanins.

- Extraction method should be thoroughly reviewed in the future, and if the search for anti-obesity active substances and the easy processing technology of food materials are developed, the consumption of mulberry leaves will help to improve income of the production farmers.
