환경성안질환은 최근 유병률이 증가하고 있는 알러지성 결막염,건성안, 백내장, 망막혈관질환으로 구분되는 질환임. 이에 대응하기 위하여 본 연구진은 치료물질로써 우리 몸의 선천면역 기전으로 존재하는 Decoy receptor D6 와 DARC를 모방하여 동시에 여러 가지 케모카인을 제거할 수 있는 디코이 트랩 (TRAP)을 제작하였음. 지역사회 코호트를 대상으로 환경성안질환에 대한 바이오마커로서 염증성 케모카인을 확인하고, 멀티케모카인 트랩에 대한 타깃을 도출해낸 후, 이를 독자적 in vivo/vitro system에서 입증하였음.

Environmental eye diseases include allergic conjunctivitis, dry eye disease, cataract disease, and vascular retinopathies. This study will attempt to (1) create a working animal model that simulates environmental eye diseases, (2) find key biomarkers and inflammatory chemokines through a community-based cohort study, (3) conduct various experiments to find inhibitory multi-chemokine traps for the aforementioned biomarkers and derive at least two such compound molecules with verified inhibitory effects, (4) determine the pharmacokinetics, pharmacodynamics, and toxicology of these compounds in preparation for clinical trials.
□ Results
1) First year: We will attempt to create a chemokine decoy trap that acts to inhibit key
biomarkers through utilization of innate immune mechanisms such as decoy receptor D6 and DARC. (2) Second year: Find that least five possible candidate compound molecules that can act as decoy traps through in vitro assay. (3) Third year: Through changes in molecular structure and chemical alterations, we will improve affinity of candidate compounds to certain chemokines and will introduce these compounds to mice models exposed to simulated environments, thereby finding at least two molecules with verified in vivo results. (4) Fourth year: determine the pharmacokinetics, pharmacodynamics, and toxicology of candidate compounds to determine metabolic rate and toxicity. (5) Fifth year: Prepare the refined candidate molecule for in vivo testing and mass production, proceed with IND filing.
□ Expected Contribution
1. Application plan
(1) Create and market a multi-chemokine trap that treat and/or prevent environmental eye diseases. (2) Investigate the pathophysiology and effects of chemokines in environmental eye diseases. (3) Utilization of chemokine traps as treatment for ocular surface diseases such as dry eye disease and allergic conjunctivitis will inhibit proliferation of memory T cell and thereby reduce incidence of those diseases. (4)Further, prospective molecules will have impact in disease that necessitate ocular surface cell chemoattraction, such as noninfectious keratitis, Steven Johnson syndrome, etc. (5) The developed multi-chemokine trap will further have systemic effects such as in immune diseases, inflammatory diseases, cancer metastasis, and organ transplantation.
2. Expected contribution
After determination of pharmacokinetics, pharmacodynamics, and toxicology of candidate, we will prepare the refined candidate molecules for in vivo testing and sound out possible relationship with pharmaceutical companies. The developed multi-chemokine trap will not only effect environmental eye diseases, but also have systemic effects such as in immune diseases, inflammatory diseases, cancer metastasis, and organ transplantation.
