본 연구의 목표는 ① 해양 은편모류 기반 핵심 원생생물을 탐색하고, ② 이들의 대량배양 시스템을 위한 원천기술을 확립하며, ③ 대량 배양된 원생생물체로부터 이차대사산물을 규명하고, 이 중 세포독성, 항산화, 소염진통 효과가 우수한 이차대사물질을 생산하기 위한 원천기술을 개발하는 것임.

연구개발내용(1000자 내외)
본 연구에서는 다음과 같은 세부목표를 설정한 후 연차별 연구를 통하여 최종 목표를 달성하고자 한다.

5. 은편모류 기반 해양 원생생물간 포식자-먹이 특이성 관계 규명
6. 대량배양 시스템 개발을 위한 최적 성장조건(환경요인 등) 규명
7. 핵심 원생생물 단종배양체의 대량배양을 통한 이차대사산물 탐색 및 규명
8. 생태생리적 특이성 발현 조건에서의 핵심 원생생물 이차대사산물 탐색 및 규명
9. 종간 포식조건에서의 핵심 원생생물 이차대사산물 탐색 및 규명
연구개발성과
본 연구팀은 연구기간 동안 은편모류 기반 해양 원생생물 80여종의 단종배양체 확립(은편모류 15종, 와편모류 50종, 섬모류 4종, 규조류 10종, 유글레나류 2종, 침편모류 1종, 녹조류 6종)과 형태 및 분자생물학적 방법을 사용하여 분류하였다. 또한, 해양 원생생물의 생태, 생리적 특성을 규명하고, 다양한 원생생물의 소규모 배양을 (와편모류 35종, 유글레나류 3종, 은편모류 3종, 남세균 2종, 녹조류 2종) 통한 생리활성을 분석하여 유용한 배양체를 선정하였다. 유용한 배양체의 대량배양을 (와편모류 4종, 은편모류 1종) 통한 독성물질, 소염진통 효능 등이 우수한 이차대사물질 분리 및 구조 규명하였으며 이 중 활성이 우수한 신규 화합물 6개를 분리하였다.본 연구 단계 동안 연구결과들은 SCI급 국제전문학술지 7편의 논문 및 22편의 국내외학술대회 논문으로 발표하였다.
활용계획 및 기대효과(500자내외)(응용분야 및 활용범위 포함)
본 연구를 통하여 얻어지는 결과는 국내 및 국제적으로 해양 원생생물을 이용한 연구는 고부가가치 물질의 탐색, 독소의 탐색, 바이오에너지개발, 유전체 확립연구, 진화모델 생물 발굴 등에 기여할 것이다. 또한, 해양 원생생물을 이용한 이러한 다양한 연구는 각각의 산업에 맞는 최적의 해양 저서조류 및 식물플랑크톤을 활용할 수 있게 할 것이므로 경제적이나 산업적으로 큰 기여를 할 것으로 기대한다.

Purpose
The major goals of this study are ① to detect cryptophyte-mediated key marine protists, ② to establish mass culture system, and ③ to explore useful secondary metabolites showing cytotoxicity, antioxidant and anti-inflammatory effect from mass-cultured strains of the key marine protists.
Purpose
The major goals of this study are ① to detect cryptophyte-mediated key marine protists, ② to establish mass culture system, and ③ to explore useful secondary metabolites showing cytotoxicity, antioxidant and anti-inflammatory effect from mass-cultured strains of the key marine protists.
Contents
To achieve the final goals, the following sub-topics of research and development should be performed during the five years:
1. Establishing clonal cultures of planktonic and benthic marine cryptophytes
2. Establishing cultures of marine cryptophyte-mediated ciliate or dinoflagellates
3. Investigating ecophysiological characteristics of marine ciliates and dinoflagellates using the laboratory cultures
4. Investigating predator-prey relationships among cryptophyte-mediated marine protist species
5. Investigating optimal growth conditions for mass cultures of marine protists
6. Exploration of secondary metabolites from the massive culture of pure species corresponding to the key protist species
7. Exploration of secondary metabolites secreted by key protist species depending on the growth conditions
8. Exploration of secondary metabolites secreted by key protist species under the stress of prey-predator relation
Development results
A total of 80 uni-protist cultures of the cryptophyte-mediated key marine protist species (15 cryptophytes, 50 dinoflgellates, 4 ciliates, 10 diatoms, 2 euglenophytes, 1 raphidophyt, 6 chlorophytes ) were set up as laboratory strains along with their taxonomy by morphological and molecular genetic studies. Small scale cultivation of the strains(35 dinoflagellates, 3 euglenoids, 3 cryptophytes, 2 cyanobacteria, 2 chlorophytes) was performed for the screening of useful strains with bioactive applicabilities. From the selected strains with useful bioactivities 4 dinoflagellates and 1 cryptophyte strains, six new secondary metabolites showing cytotoxicity, and anti-inflammatory effect were isolated from their massive cultures, and were followed by the structure determination. Seven SCI papers and 22 scientific presentations in were resulted from the 1st step R&D work.
Expected Contribution
1. The results of this study would contribute to the detection and development of novel metabolites, toxin, bioenergy, genomic materials, and model organism for evolution research.
2. The results of this study would be highly beneficial in future use in economic and industrial aspects.
