□ 연구결과 [1차년도] LPS 및 OGD/R 노출된 BV2 소교세포에서 BMP/pSmad 신호경로 증가와 염증성 사이토카인 발현, 철대사조절단백인 hepcidin/ferroportin 증가를 확인하고, noggin으로 BMP 신호경로 봉쇄시 M2 및 철유리형 소교세포로 표현형으로 전환되는 것을 확인함. 이를 통해 BMP 신호경로 변경을 통한 소교세포 표현형 조절이 가능함을 규명함. [2차년도] 젊은 성체 마우스에 MCAO 유도 2주후 noggin(1μg/day)을 2주간 동측 뇌실내 투여, 1주 간격으로 행동학적 실험을 진행하고 8주에 뇌용적을 측정하였을 때, neurological score, hanging wire test 가 대조군에 비해 호전되고 뇌위축 정도가 감소함을 확인하였고, 뇌세포내 철대사의 변화 및 항염증성 사이토카인 증가, 재수초화 증가를 확인함. 이를 통하여 1차년도 결과와 더불어 허혈 손상 후 만성기의 BMP 신호 조절로 M2 세포의 수복 작용 증가를 통해 신경학적 증상이 호전 가능함을 규명함. 또한 젊은 성체 대비 20개월령 노령 마우스의 전신 혈액내 염증성 사이토카인 증가 및 뇌내 M1 마커와 철 증가를 확인하여 뇌노화 과정에 염증성 반응 증가를 확인함. [3차년도] 노령 마우스의 증가된 내장지방을 제거 후 4주째 측정한 전신 및 뇌내 염증반응이 대조군에 비해 감소하였고, 이들을 MCAO 유도 후 3일째 뇌경색 용적 측정시 대조군에 비해 뇌손상이 감소하는 것을 확인하여, 이를 통하여 노화에 따른 내장지방 증가가 뇌노화및 뇌손상 증가에 밀접한 관련이 있음을 증명함. 또다른 실험으로 노령 마우스에 복강내 noggin(0.5 mg/kg)을 1주일간 주입했을 때 염증반응이 감소하고, MCAO 유도 후 8주간 행동학적 실험한 결과, 대조군에 비해 사망률 감소 및 신경학적 기능 호전을 관찰함. 이러한 결과는, 뇌에 직접적으로 작용하는 약물뿐만 아니라, 노화와 동반된 말초 염증을 조절하는 것도 노인성 뇌질환 치료에 도움이 되는 것을 보여줌.

□ Purpose& contents This study was aimed to define the effects of BMP signaling on microglia/macrophage phenotypes, neurological outcomes and neurorestoration durin chronic phase of ischemic stroke, and also to define the effects of macrophage phenotypes and inflammatory responses in the increased visceral adipose tissues with aging on neurorestoration using aged mice. The final goal of the study is to suggest a therapeutic strategy of neurorestoration by modulation of microglia/macrophage phenotypes in the ischemic brain during chronic phase through defining factors involved in repair process. The study was composed for 3 years. The effects of BMP signaling on microglial iron metabolism and phenotypes (1st year), the effects of BMP inhibition on ischemic brain repair process via M2 phenotype cells and M1/M2 phenotype changes in the aged brain (2nd year), and the effects of visceral fat with aging on M1/M2 and ischemic brain repair (3rd year) were defined.
□ Result [The 1st year] Activation of BMP/pSmad signaling, the increase of inflammatory cytokines and hepcidin/ferroportin were blocked by noggin, and nogging shifted microglial phenotype to M2 with iron release under LPS or OGD/R, suggesting the shift of microglial phenotypes by modulation of BMP signaling. [The 2nd year] Neurological outcomes and brain atrophy were improved in young adult mice treated with intraventricular infusion of noggin (1 μg/day) for 2 weeks after ischemic stroke, and their brain showed alteration of iron metabolism, the increase of anti-inflammatory cytokines and remyelination. The results combined with those of the 1st year suggest that the improvement of neurological outcomes can be possible through the increase of tissue repair processes mediated by M2 cells via BMP inhibition during chronic phase of ischemic stroke. Also, the increase of systemic inflammatory cytokines, and the increase of M1 markers and the iron level were observed in the brain of aged mice (20 months) compared with young mice, implying increasing inflammatory responses in the aged brain. [The 3rd year] Systemic and brain inflammatory responses were decreased, and ischemic brain injury induced by MCAO with reperfusion for 3 days was also decreased in the aged mice which were subjected to surgery of visceral fat removal 4 weeks before. The results show that the increase of visceral fat with aging is closely associated with brain aging process and the increase of brain injury. In another set of experiment, aged mice treated with noggin (0.5 mg/kg, i.p.) for 1 week showed less inflammatory responses in visceral fat, and showed less mortality with better neurological outcomes 8 weeks after MCAO. The results indicate that controling peripheral inflammation associated with aging is helpful to treat brain diseases with aging, in addition to drugs direcely acting in the brain.
