□ 연구의 목적 및 내용
본 연구에서는 고감도 및 고선택적인 칩 기반의 센싱 기술(예: 나노바이오 및 소프트 계면 융합형)을 개발하여 수질 오염원 항생제 의약품의 분석 및 지속적인 모니터링에 활용할 뿐 아니라 기타 신종 의약품과 오염원 분석에 적용함을 최종 목표로 두고 있다. 이를 위해 우수한 광-전기 기능성의 바이오나노 복합체 합성, 타겟 선택적인 바이오리셉터 페어 고안, 표면 샌드위치 분석 기술 및 소프트 계면 기반 검출의 원천 보유 기술들을 융합하여 항생제 수질 오염원 맞춤형 센서로 전환하고 실시간으로 신속하게 검출이 가
□ 연구의 목적 및 내용

□ 연구결과
본 연구는 수환경 내 잔존하는 의약품의 농도와 양을 실시간으로 모니터링하기 위한 고선택적 및 고감도의 나노입자융합형 바이오센서를 개발하는 것으로 총 연구기간 동안 특히 인체와 동물에 주로 사용되어왔던 수질 오염원 항생제(예: 테트라사이클린(tetracycline), 옥시테트라사이클린(oxytetracycline), 반코마이신(vancomycin), 카나마이신(kanamycin), 독소루비신(doxorubicin), 세포티암(cefotiam), 레보플록사신(levofloxacin), 로메플록사신(lomefloxacin)를 주요 타겟으로 정하고, 항생제 맞춤형의 센싱 플랫폼을 개발하였다. 1차년도에는 한 개의 타겟 물질당 두 개의 리간드(압타머와 항체)를 활용하는 새로운 표면 샌드위치 플랫폼의 바이오센서를 고안하여 센서의 선택성을 증폭시키는 노력을 하였다. 표면 플라즈몬 공명 분석법을 이용하여 개발한 각 리간드와 항생제간의 결합력을 확인하였으며, 검증된 리간드 페어중 한 개를 표면에 고정하고, 다른 한 개의 리간드와 샘플을 센서 표면에 흘려주어 표면 샌드위치 복합체를 형성하였으며 전기화학 및 광학 기반의 표면 민감형의 센싱 플랫폼에 융합하여 항생제의 농도를 분석하였다. 이와 함께 개발한 샌드위치 센서의 감도를 증대하기 위해 광학적 및 전기화학적으로 우수한 특성을 갖는 금 및 나노입자에 리간드 중 한 개를 고정하여 센서에 융합시켰으며, 이를 통해 센서의 감도를 약 천배에서 십만배 이상 향상시켜 물 시료내 에 존재하는 극미량(수십 ppt) 항생제 분석이 가능함을 증명하였다. 또한 항생제 약물의 지질친화도 정도 및 pH에 따라 전하가 달라지는 특성 등을 정확히 이해하여 좀 더 성능이 우수한 센서를 개발하기 위해 편극화된 소프트 (액체/액체) 계면에서 약물의 전이반응 연구도 함께 수행하였다. 최종적으로 개발된 바이오센서의 진단킷 및 소형화와 함께, 실제 샘플 적용 시 검출방해효과를 제거하기 위한 표면화학을 고안하여 성능을 최적화하였으며, 의약품 환경오염원 맞춤형의 정량분석법을 확립하였다.
□ 연구결과의 활용계획
● 독창적인 나노소재물질의 설계 기술 및 신규 바이오센서 개척이라는 탁월한 연구 성과를 통해 기초과학 분야에서 기술선진국을 능가할 수 있는 연구기반 마련.
● 칩 기반 센싱 기술은 다양한 종류의 타겟 탐침용 리셉터의 도입이 용이하므로 항생제 수질오염원뿐 아니라 이와 관련된 신종 의약품과 기타 다양한 오염원 분석에도 확대 응용이 가능.

The goal of the proposed research is to establish a highly selective and sensitive chip based sensing platform for continuous monitoring and detection of antibiotics in addition to newly developed pharmaceuticals for water quality control. This will be achieved by combining the synthesis of nanobio conjugates, surface sandwich assays, new bioreceptors targeting antibiotics alongside soft interface based sensing platforms. Extensive efforts will also be accompanied to integrate these core techniques to develop antibiotic selective sensing platforms with real-time, fast, and accurate analysis capabilities.
□ Result
Our research work was focused on the development of highly selective and sensitive nano integrating biosensors for continuous monitoring of water polluting pharmaceutical reagents. During the grating period, we developed antibiotic selective sensing platforms particularly targeting for tetracycline, oxytetracycline, kanamycin, vancomycin, levofloxacin, doxorubicin, cefotiam, lomefloxacin which have been widely used for both human and animals. For the first granting year, novel surface sandwich bioassay involving a pair of ligands for one target to enhance the selectivity of sensing methods. Real-time surface plasmon resonance technique were used to select the set of ligand pair and target antibiotics showing the strongest binding affinity towards each other and also to confirm the chosen ligand pair having different binding sites to the target to form sandwich complexes. The surface sandwich formation methods were applied to both electrochemical and optical sensing platforms to quantitatively analyze target molecules. In addition, ligand molecule functionalized gold, silver and carbon nanoparticles with excellent optical and electrical properties were implemented onto both sensing platforms to improve the sensitivity measuring about ppt concentrations of antibiotics. This is at least 1,000 to 100,000 times improvement when compared to surface sandwich assays without any nanoparticles involved. In order to understand the lipophilicity and charge properties of target drug molecules at different pH buffered solution, ion transfer reactions across a ploarized soft (liquid/liquid) interface were investigated using voltammetry. Also, the developed sensors were transformed to a disposable real time sensing platform using screen printing and microfab techniques in addition to developing surface modification chemistry to reduce non-specific interfering adsorption effect from water samples. Finally we established new quantitative analytical methodologies for environmentally toxic pharmaceutical reagents.
