□ 연구의 목적 및 내용
연구목적
골다공증과 연계된 질환 (폐경, 염증성, 다발성 골수종)에서 높은 발현 패턴을 보이는 셀레노 단백질 W (selenoprotein W:Sepw1) 유전자의 골 제어 기전연구와 이의 역할을 규명하고자함
연구내용
1. Sepw1 유전자 적중 (knock-out: KO) 및 형질전환 (transgenic: TG) 마우스의 골 분석
1) 골 지표인자 분석 (μCT 및 골 조직 절편)
2) 파골세포 분화 (성장, 부착, 이동, 융합) 및 골 흡수 평가

□ 연구의 목적 및 내용
연구목적
골다공증과 연계된 질환 (폐경, 염증성, 다발성 골수종)에서 높은 발현 패턴을 보이는 셀레노 단백질 W (selenoprotein W:Sepw1) 유전자의 골 제어 기전연구와 이의 역할을 규명하고자함
연구내용
1. Sepw1 유전자 적중 (knock-out: KO) 및 형질전환 (transgenic: TG) 마우스의 골 분석
1) 골 지표인자 분석 (μCT 및 골 조직 절편)
2) 파골세포 분화 (성장, 부착, 이동, 융합) 및 골 흡수 평가
3) 파골세포 관련 세포신호전달 분석
2. Sepw1 KO/TG 마우스에 난소적출 골다공증 모델 형성 후 골 분석
1) 골 지표인자 분석 (μCT 및 골 조직 절편)
2) 대조군과 비교하여 Sepw1 과발현에 의한 골다공증 증감 유무를 확인 (Sepw1 발현 정도와 골다공증 연계성 판단)
3. Sepw1 유전자 발현과 다발성 골수종 환자 (multiple myeloma)의 골다공증 연계성 확인
1) IRB 신청 후 다발성 골수종 환자 혈액샘플 확보
2) 파골 전구세포인 PBMCs (peripheral blood mononuclear cells)는 Ficoll (Histopaque) gradient를 이용하여 분리
3) 파골세포 분화 (성장, 부착, 이동, 융합) 및 골 흡수 평가
4) 파골세포 관련 세포신호전달 분석
□ 연구개발성과
1. Sepw1 유전자 적중 및 형질전환 마우스를 이용한 Sepw1의 골 질환 연계성 확보
- Sepw1 유전자 적중 마무스는 골화석증 병변을 보임
- Sepw1 유전자 형질전환 마우스는 골다공증 병변을 보임
2. Sepw1 유전자와 골 관련 질환 (염증성, 폐경, 다발성 골수종)과의 연계성 규명
-염증성, 폐경, 다발성 골수종 질환 모델에서 Sepw1 유전자의 높은 발현패턴 보임
-Sepw1 유전자의 발현증가는 골 질환과 연계성 있음
□ 연구개발성과의 활용계획 (기대효과)
1) 필수미량원소로 알려진 셀레늄을 함유한 셀레노 단백질의 새로운 기능 제시로 셀레늄 관련 학문에 활용될 수 있을 것으로 기대됨
2) Sepw1 유전자 발현 조절에 따른 골 질환을 제어할 수 있는 새로운 표적인자로 Sepw1 유전자를 제시할 수 있을 것으로 기대됨
3) 본 과제에서 제시하는 골다공증 모델 (폐경, 염증성, 다발성 골수종) 이외의 골다공증 질환 (노화, 비만, 골 전이성 암, 류마티스 관절염 등)에서 Sepw1 유전자 발현과 질환 연계성 규명에 활용될 수 있음
