□ Purpose&Contents
The purpose of this study is the development of improvement of technology honeybee valuable gene-based disease resistance character, consisting of four sub-tasks.
1. Development of disease control and long-term preservation technology based on useful genes of bees
- Investigation and utilization of the molecular mechanisms of honeybees response to pathogens
- Discovery of genetic resources for bioactive substances related to maintenance of excellent lineage bees
2. Selection and development of substitute compounds for Fumagillin to control Nosema disease
- Screening of potent alternatives for fumagillin, the only effective antimicrobial agent currently using againstmicrosporidial parasite N. ceranae, was performed using natural and chemically synthesized compounds
- The compounds were subjected to evaluation of anti-nosemosis activity
3. Transcriptome analysis of disease-resistant bees and development of technology for freezing and preserving genetic resources
1) Transcriptome analysis of disease-resistant bees
2) Development of semen freezing technology
4. Application and commercialization of substances useful for managing honeybee diseases
□ Results
1. Development of disease control and long-term preservation technology based on useful genes of bees
- MRJP4-C functions as an effective antimicrobial agent that exhibits activity against bacteria, fungi, and yeast.
- An increased expression of antioxidant proteins could facilitate prolonged storage and survival of sperms in the spermatheca of mated queens, suggesting the role of antioxidant proteins in antioxidative defense against ROS.
2. Selection and development of substitute compounds for Fumagillin to control Nosema disease
- Eight compounds were purified and isolated from the MeOH extract of Cordyceps militaris Media.
- Twelve compounds were isolated from the snowbell (Styrax japonica) honey.
- Four phenolic compounds [3-pentylcatechol (PC), 4-pentylgallol PG), malaxinic acid (MA), MA algycone] were chemically synthesized.
- In addition, total 28 compounds including 21 compounds of phenolics and monoterpenes were subjected to evaluation of anti-nosemosis activity.
- Finally, gallol and PC showed significantly higher activities than fumagillin and PG exhibited activity in similar level to fumagillin.
- That is, these three compounds (gallol, PC, PG) were selected as potential benefit alternatives for fumagillin.
3. Transcriptome analysis of disease-resistant bees and development of technology for freezing and preserving genetic resources
- Through the analysis of the transcriptome of disease-resistant and non-resistant bee strains, it was confirmed that many genes related to molecular function, biological function, and pathway were expressed, and genes related to defense (Camata, Pxd, mtm, Spla2, Wrapper, Cad89D, Nurf-38) show strong resistance to virus.
- Disease-resistant strains contain lots of anti-viral RNA fragments in royal jelly, applicable to a new breed identification marker.
4. Application and commercialization of substances useful for managing honeybee diseases
- An expression analysis of the functional AMP5 peptide derived from honeybees identifies immune-related genes expressed in epidermis, fat and poisonous glands
- Development of diagnostic assay based on the poalr tube protein 3 (ptp3) expression level
□ Expected Contribution
1. Development of disease control and long-term preservation technology based on useful genes of bees
- Development of new management technology using genetic resources to control honeybee disease and pests and this development can contribute to stable productivity improvementof domestic beekeeping industry
- Fostering excellent varieties through establishment of excellent techniques for long-term preservation of bee sperm
2. Selection and development of substitute compounds for Fumagillin to control Nosema disease
- The finally selected 3 compounds are remarkably different from fumagillin in chemical structure. Therefore, it may trigger the further extensive research on the biological mechanism for control and prevention of nosemosis.
- It is expected that the three compounds could be used as fumagillin alternatives.
3. Transcriptome analysis of disease-resistant bees and development of technology for freezing and preserving genetic resources
- Contributing to the development of the beekeeping industry by developing diagnostic markers for disease-resistant strains
4. Application and commercialization of substances useful for managing honeybee diseases
- Study on the major disease control technology of honeybees using genes and natural compounds
- Development of diagnostic methods for selecting candidates for anti-Nosema compounds
