□ Purpose
The purpose of this project is to develop a source technology of nanobiocatalytic materials that can achieve high stability of enzymes. This technology can be a basis for the development of nanobiocatalytic filters and 3D-structures for the prevention and removal of microbial contamination, respectively. Another goal is to develop the source technology of cell-binding domains, which can realize selective microbial killing and detection.
□ contents
Under collaborative researches with Foreign Research Laboratory (FRL), we have developed various nanobiocatalytic materials with high enzyme loadings and stabilities by deeply understanding the interactions between enzymes and nanostructured materials. Platform technologies were developed to maximize the applicability of various nanobiocatalytic materials in the prevention and removal of microbial contamination. In particular, we could develop a source technology for the convenient removal of microbial contamination by combining 3D-printed structures and nanobiocatalytic materials. Finally, we have performed in-depth researches on cell-binding domains, enabling the selective binding of cell-lytic enzymes, and could develop new technologies for selective microbial sterilization and detection using those cell binding domains.
Qualitative goals and outcomes
- Development and performance evaluation of nanobiocatalytic materials for microbial decontamination: Based on deep understanding of interactions between enzymes and nanostructured materials, highly-stabilized nanobiocatalytic materials with high enzyme loadings were developed for microbial decontamination.
- Development of eco-friendly microorganism-killing technology using nanobiocatalytic materials: To maximize the applicability of nanobiocatalytic materials, we developed nanobiocatalytic 3D-structures by combining nanobiocatalytic materials and 3D-printed structures for efficient and eco-friendly killing of microorganisms.
- Development and performance evaluation of nanobiocatalytic filters for antifouling: By applying nanobiocatalytic materials of antifouling enzymes to filters (membranes), nanobiocatalytic filters with high enzyme loading and stability could be developed, and their antifouling effect could be successfully evaluated.
- Development of cell-binding domain technology: Basic researches were performed on cell-binding domains that can selectively bind onto microorganisms as a constituent of cell wall-lytic enzyme. By effectively combining these cell-binding domains with various nanostructured materials, we could develop technologies for selective microbial killing and detection.

Quantitative goals and outcomes
In total project period, 42 (10 co-papers) paper publications, 31 patents and 4 technology transfers were achieved.
- Phase 1: 18 papers (goal of 12), 5 co-papers (goal of 3.6), 5 patents (goal of 1), 2 technology transfers (goal of 0)
- Phase 2: 24 papers (goal of 12), 5 co-papers (goal of 4.8), 26 patents (goal of 3), 2 technology transfers (goal of 0)
□ Expected Contribution
Nanobiocatalytic materials and their complexes with filters and 3D-printed structures, developed in this project, are anticipated to find their applications not only in microbial decontamination, but also in various other fields such as bioconversion, biosensors, biofuel cells, and carbon dioxide capture/conversion/utilization. Cell-binding domains, capable of selectively binding onto microbial cell walls like antibodies, are expected to play a role of antibodies in the fields of microbial diagnosis and sterilization. The production of cell-binding domains using E. coli would be one of the important advantages in their competition with antibodies.
