1. 라이프케어 소재용 두류 유용 돌연변이 유전자원 개발
1-1. 방사선 후대 대두 유전자원 파이토케미컬 고함유 유전자원 선발
가. 콩 돌연변이 유전자원 이용 불포화지방산 함량 변이 계통 선발
- 단백콩, 대풍콩 유래 변이 계통의 불포화지방산 변이 분석
- 감마선 조사를 통한 단백, 대원 유래 돌연변이 집단 구축
- 농업형질이 우수한 계통 선발 후, 리놀렌산 함량 증가된 10개 계통의 선발

Ⅳ. 연구개발결과
1. 라이프케어 소재용 두류 유용 돌연변이 유전자원 개발
1-1. 방사선 후대 대두 유전자원 파이토케미컬 고함유 유전자원 선발
가. 콩 돌연변이 유전자원 이용 불포화지방산 함량 변이 계통 선발
- 단백콩, 대풍콩 유래 변이 계통의 불포화지방산 변이 분석
- 감마선 조사를 통한 단백, 대원 유래 돌연변이 집단 구축
- 농업형질이 우수한 계통 선발 후, 리놀렌산 함량 증가된 10개 계통의 선발
- 단백콩 유래 계통인 DB-HL4, 5의 경우 원품종인 단백콩의 리놀렌산 함량(6.53%) 보다 약 1.7배 증가된 11.29% (HL4), 10.88% (HL5) 함량 증대를 보였고, 대풍콩 유래 계통인 DP-HL6의 경우도 원품종인 대풍콩의 리놀렌산 함량 (7.15%) 보다 최대 약 1.6배 증대된 리놀렌산 함량을 나타냄.
- 선발된 10개 계통의 리놀렌산 함량 변화 관련 유전자 탐색 연구 수행
나. 올리고당 함량 변이 대두 계통 선발
- 감마선 조사를 통한 팔달 유래 돌연변이 집단구축 육성
- HPLC로 각 계통 종자의 전체 당 성분 중 sucrose 함량과 올리고당인 raffinose, stachyose 함량을 분석한 결과, 선발된 전체 계통에서 전체 당 함량이 증가하였고 그 중 I01 계통이 11.81 mg/100g 으로 가장 높은 함량을 나타냄
다. 비린내 저감 돌연변이 콩 신품종 청미원 개발 및 품종보호권 출원
- 원품종 황금콩 종자에 방사선육종을 이용하여 비린내가 제거된 우수 계통 (H-70)을 선발, 청미원으로 명명하여 품종을 출원함(출원번호: 2018-118)
1-2. 파이토케미컬 고함유 대두 계통 특성 분석 및 유전체 변이 탐색
가. 파이토케미컬 고함유 대두 계통 품종화 기초 데이터 분석
- 원품종 포함 총 210 계통의 MDP(Mutant Diversity Pool) 구축
- 기초 농업형질 데이터 확보(꽃색, 배축색, 신육형, 경장, 마디수, 분지수, 종피색, 배꼽색과 개화일수, 성숙일수, 백립중)
- TRAP 마커를 활용한 대두 MDP 유전적 다양성 규명
나. 대두 지방산 성분 변화 관련 유전자 변이 탐색 및 발현 수준 확인
- 총 10개 리놀렌산 함량 변화 돌연변이체의 유전자 수준 차이 규명
- FAD2 유전자 6개, FAD3 유전자 4개 염기서열 확보(FAD2-1A (2,151bp), FAD2-1B (1,830bp), FAD2-2A (1,409bp), FAD2-2B (2,112bp), FAD2-2C (2,092bp), FAD2-2D (2,088bp), FAD3A (4,250bp), FAD3B (4,104bp), FAD3C (2,724bp), FAD3D (3,431bp))
- 염기서열 기준 품종인 william82와 비교시 FAD2-1A에서 4개, FAD2-1B에서 4개, FAD2-2A에서 5개, FAD2-2B에서 1개, FAD2-2C에서 3개, FAD3B 에서 18개의 SNP가 확인 되었으며, 특히, FAD2-2C에서는 DB-HL3 개체에서 15bp의 insertion이 확인됨
- 대조구 등 비교를 위하여 4개의 대두 종자형성 developmental stage에서의 각 유전자별 발현 양상 비교한 결과 stage 2, 3 단계에서 특이적으로 발현값이 원품종에 비해 증가함을 확인
- stage 3에서 FAD3C, FAD3D 유전자의 돌연변이체 발현양의 증가로 인하여 리놀렌산 함량 증대에 영향 확인함(논문 심사 중)
1-3. 대사성질환 개선용 lentil, L-Dopa 고함유 faba bean 유전자원 선발 및 변이유기
가. Lentil 수집종의 유전자원 선발 및 세대 진전
- 총 26개 전 세계 수집 유전자원 중 국내 환경 적응 및 농업형질 우수 자원 6계통 선발
- 감마선 감수성 평가를 통한 최적 선량 확보를 통한 렌틸 방사선육종 전개 개시(논문 게재 완료)
- 2017년 계통 당 control 포함 2,000립, 총 12,000 M1 식물체 포장전개 및 M2 종자 pooling
- 2018년 총 2,827개체 M2 세대 진전 및 개체 수확
- 2019년 우수형질 선발을 통한 M3 세대진전 예정
- 2018년 추파를 통한 국내 월동 능력 검정
나. Faba bean 수집종의 유전자원 선발 및 세대 진전
- 총 371개 전 세계 수집 유전자원 중 국내 환경 적응, 농업형질, 동계생존율
우수 자원 10계통 선발(논문 게재 완료)
- 감마선 감수성 평가를 통한 최적 선량 확보를 통한 faba bean 방사선육종 전개 개시
- 2017년 계통 당 control 포함 250립, 총 2,500 M1 식물체 포장전개 및 M2 종자 pooling
- 각각의 mutant line 별로 30%~84% 월동율 확인(2018년 1월 정읍 최저 온도 –14℃)
- 2018년 10월 추파를 통한 총 3,850개체 M2 세대 진전 중
- 2019년 우수형질 선발을 통한 M3 세대진전 예정
1-4. 유용돌연변이 자원 선발 시스템 구축 및 성분 특성 분석
가. 분자마커시스템 구축 및 돌연변이율 검정
- TRAP 마커 시스템을 활용한 552개 M1 식물체 DNA의 돌연변이율 검정
- 감마선 선량과 genotyping의 correlation 확인(논문 심사 중)
나. 두류의 기능성 물질 분석 및 성분 활성 분석
- HPLC, GC-MS를 이용한 faba bean 부위별 L-Dopa, fatty acids, 및 isoflavone 함량 비교 분석 완료(논문 게재 완료)
- Faba bean 전분의 물리화학적 특성 분석 결과 타 곡류들과 달리 30℃에서도 고온에서의 용해도와 유사한 수준으로 높게 나타남, 신규 전분 category 가능성 높음
- 대두 MDP line 중 원품종 단백의 변이 라인(D-28-01)에서 isoflavone 함량이 원품종 대비 5.5배 가량 크게 증가한 계통 확인
- 단백, 대풍, 팔달 품종에서 총 플라보노이드 함량이 증가한 10개 계통의 변이체 확인
- 항치매는 faba bean, lentil, soybean에서 유사한 활성
- 항미백은 lentil과 soybean에 비해 faba bean에서 높은 활성을 보임
- 항산화는 lentil에서 soybean과 faba bean 보다 월등히 높은 활성을 확인함
다. 기능성 두류 파이토케미컬 최적 이용을 위한 가공적성 및 성분변화 연구
- Faba bean의 가공처리에 따른 기능성 활성 및 물질 분석 실험 완료
- Lentil 3가지 품종의 가공처리 전후 기능성 변화 검정 완료
라. Faba bean 내한성 관련 유전자 탐색
- PAC-Bio를 이용한 Faba bean 돌연변이 계통 발현유전체 비교 분석 완료
- 총 6개의 내한성 유전자 후보군을 대상으로 96개 contig 유전자 정보를 획득, 향후 qRT-PCR을 통해 실제 저온 스트레스 시 발현 양상 확인 예정
2. 방사선육종 식물자원 활용 기능성 효능 검증 및 실용화 기반연구
2-1. 방사선육종 아피오스로부터 대사체 분리 및 생리활성 분석
가. 방사선육종 아피오스로부터 대사체 분리 및 구조분석
○ 방사선육종 아피오스 용매별 분획물 획득
- 에틸아세테이트와 부탄올 분획물에서 총 20개의 대사체를 분리 완료
- NMR 분석을 통해 구조분석 완료
나. 방사선육종 아피오스 대사체의 생리활성 평가(논문 게재 완료)
○ 방사선육종 아피오스 대사체의 항염증 효능 평가
- 20개의 대사체 중 4개(AP13, AP7, AP14, AP58)가 항염증 효능을 보였음
○ 방사선육종 아피오스 대사체의 대사성질환 효능 평가(논문 심사 중)
- 20개의 대사체 중 2개(AP2, AP7)가 α-glucosidase 효소 활성을 억제하였음
- AP7의 경우 50 μM 농도에서 DPP-IV 효소 활성을 50% 저해하였음
다. 방사선육종 아피오스에서 분리된 lupinalbin A의 항염증 효능 기작 규명(논문게재 완료)
○ LPS로 자극된 RAW264.7 세포에서 lupinalbin A의 항염증 효능 기작 규명
- Lupinalbin A가 농도 의존적으로 iNOS의 mRNA와 단백질 발현을 억제하였음
- Lupinalbin A가 농도 의존적으로 IL-6, TNF-α, MCP-1의 생성을 억제하였음
- Lupinalbin A가 IFN-β의 생성과 STAT1 단백질 발현을 억제하였음
2-2. 방사선육종 두류 추출물 확보 및 생리활성 평가
가. 방사선육종 두류 150계통 추출물 제조 및 LC/MS 분석 조건 확립
○ 11종의 standard compounds을 이용해 UPLC/ELSD 및 UPLC/Q-tof MS 장비에 대한 최적의 분석조건을 확립
○ 단백콩 샘플을 이용해 LC/MS 분석 전 전처리 조건 확립
나. 방사선육종 단백콩 45 계통 최적 추출물에 대한 LC/MSMS 분석 및 분자네트워킹 조건 확립
○ 11종의 standard compounds을 이용하여 구조적 유사성에 따라 집단화 (clustering)를 수행하는 최적의 분자네트워킹 분석 조건 확립
- 단백콩 45종 추출물 및 11종의 standard compounds MS/MS 분석결과를 토대로 추출물 내 대사물질의 구조적 정보를 확인함
다. 다변량 통계기법
○ PCA, PLS-DA와 HCA의 방법을 수행하여 원품종 단백콩과 방사선 육종 단백콩 간의 차이를 규명함
라. 방사선육종 두류 150계통 추출물의 대사성질환 생리활성 평가
○ 방사선육종 두류 150계통 추출물을 이용하여 alpha-glucosidase와 DPP4 효소활성을 억제하는지 여부를 평가하였음
- 효소활성을 평가한 결과 방사선육종 두류 150계통 중 억제효능이 우수한 계통을 선발하지 못하였음
2-3. 방사선육종 렌틸 추출물 확보 및 생리활성 평가
가. 방사선육종 렌틸 추출물 제조 및 성분 분리
○ 렌틸을 이용하여 에탄올 추출물 및 용매별 분획물 획득 완료
○ 렌틸의 부탄올 분획물로부터 대사체 성분 5개를 분리 및 구조분석을 완료하였음
나. 렌틸 추출물 및 성분에 대한 생리활성 분석
○ TBK-1 효소 활성 억제능 평가
- 렌틸의 부탄올 분획물에서 분리된 5개 중 4개의 화합물이 농도 의존적으로 TBK-1 활성을 억제하였음
- 렌틸의 에탄올 추출물과 용매별 분획물을 이용하여 TBK-1 효소 활성 억제능을 평가한 결과 억제 효능을 확인하지 못함
○ 대사성질환 효능 평가
- 렌틸의 부탄올 분획물에서 분리된 3개의 화합물을 이용해 alpha-glucosidase와 DPP-IV 효소활성을 평가한 결과 3개의 화합물 모두 농도 의존적으로 두 효소의 활성을 억제함을 확인하였음
다. Molecular docking 분석(논문 게재 완료)
○ Kaempferol의 저혈당 메커니즘 작용에 근거하여 Kaempferol 유도체와 DPP-IV 사이의 가능한 결합 상호작용을 조사하였음
- DPP-IV 효소활성을 억제하는 3개의 화합물 모두 효소의 S1과 S2의 넓은 공간에 잘 맞았으며 모든 flavone core 구조는 주로 S1에 위치하고 이들의 glycoside는 S2의 잔기와 상호 작용함
2-4. 방사선육종 차조기 포함 추출복합물을 이용한 관절염 개선 기능성식품 개발에 대한 후속 연구
가. 방사선육종 차조기와 원품종 차조기의 초임계 추출물 제조 및 성분 분리(논문 게재 완료)
○ 방사선육종 차조기와 원품종 차조기의 건조된 잎으로서 각각 초임계 추출물을 획득
○ 방사선육종 차조기 추출물로부터 9-hydroxy isoegomaketone (신규 화합물)과 원품종 차조기 추출물로부터 myristicin을 분리 및 구조분석 완료
○ 방사선육종 차조기와 원품종 차조기 추출물로부터 4개의 monoterpene 화합물의 함량을 비교분석 하였음 나. 신규화합물 (1)의 항염증 생리활성 분석 연구
○ LPS로 자극된 RAW264.7 세포에서 신규 화합물은 농도 의존적으로 염증인자의 생성을 억제하였음
다. 방사선육종 차조기 추출복합물을 이용한 만성질환동물에서 관절염 개선 효능 평가(논문 게재 완료)
○ 관절염 만성질환 동물모델에서 방사선육종 차조기 추출복합물의 효능 평가
- Clinical arthritis scoring 및 부종 두께 모두 방사선육종 차조기 추출복합물을 투여한 군에서 유의적으로 감소하였음
- 혈청 내 염증인자의 경우 IL-1beta의 생성이 방사선육종 차조기 추출복합물을 투여한 군에서 유의적으로 감소하였음
- 방사선육종 차조기 추출복합물 133.0 mg/kg/day를 투여한 군에서 항관절염 예방 효력이 있음을 확인하였음
라. 방사선육종 차조기 추출복합물의 단회 경구 투여 독성시험 수행
○ 차조기 추출물의 개략의 치사량은 암컷의 경우 1,000 mg/kg 전후이며, 수컷의 경우 1,000 mg/kg과 2,000 mg/kg사이로 확인되었음.
마. 방사선육종 차조기 추출복합물의 14일 반복 경구 투여 독성시험 수행
○ 차조기 추출물 최대내성용량은 수컷은 300 mg/kg/day 이상, 암컷은 150mg/kg/day 이상으로 사료되었으며, 장기 중량 및 부검소견에서 흉선 및 비장의 변화가 관찰된 것으로 보아 target organ은 흉선 및 비장으로 사료되었음
바. 방사선육종 차조기 추출복합물의 기술이전 실시
○ 기술이전 계약 체결 완료
- SFC바이오, 2018년 8월 28일
- 정액기술료: 30,000천원, 마일스톤: 170,000천원, 경상기술료: 매출액 2%
2-5. 방사선육종 차조기로부터 분리된 신규화합물 대량 분리법 개발
가. Centrifugal partition chromatography (CPC)를 이용한 신규 화합물 분리 (논문 게재 승인 완료)
○ CPC를 이용해 방사선육종 초임계 추출물에서 신규 화합물 9-hydroxyisoegomaketone
(1) 과 지표 물질인 isoegomaketone (2), Perillaketone (3) 세 물질을 동시에 대량 분리 하고자 함
나. CPC 분리 조건 확립을 위한 분배계수 (K) 및 분리 계수 (α) 확인 및 용매 시스템 결정
○ 다양한 용매 조건으로 K 와 α 값을 확인 하였고, n-hexane/Ethyl acetate/ethanol/water 용매 시스템으로 descending mode 에서 5:5:5:5, ascending mode 에서 8:2:8:2 조건이 3가지 지표물질을 분리 하는데 가장 적합함
다. CPC를 이용한 지표물질 분리
○ Descending mode를 이용하여 Ⅰ (2.4mg, , 96.7% purity) 분리하였으며, 휘발성이 큰 Ⅱ(56.1mg, 97.6% purity), Ⅲ (78.6mg, , 96.1% purity) 지표 물질은 ascending mode를 이용하여 분리함
2-6. 방사선육종 국화 품종을 이용한 항산화 기능성식품 개발 후속 연구
가. 방사선육종 국화 (아티다크초콜렛), 원품종 국화 (노블와인), 생약 국화 추출물 및 분획물 제조 및 항산화 활성 평가
○ 아티다크초콜렛, 노블와인, 생약에 대해 각각 에탄올 추출물 획득
○ 아티다크초콜렛 에탄올 추출물로부터 용매별 분획물 획득
○ DPPH와 ABTS 방법을 이용해 항산화 활성을 평가한 결과 아티다크초콜렛이 가장 높은 활성을 보임
다. 방사선육종 국화 (아티다크초콜렛) 성분 분리
○ 아티다크초콜렛의 에틸아세테이트 분획물로부터 7종의 성분을 분리하여 구조 분석을 완료함
라. 방사선육종 국화 (아티다크초콜렛), 원품종 국화 (노블와인), 생약 국화 추출물의 함유성분 비교 분석(논문 게재 완료)
○ 아티다크초콜렛이 노블와인과 생약에 비해 플라보노이드 배당체 및 페놀성 화합물 함량이 높았으며 반대로 플라보노이드 아글리콘 함량은 낮았음
2-7. 방사선육종 국화 품종 (아티다크초콜렛)과 시판중인 국화차 티백의 침출 온도 및 시간에 따른 비교 연구
가. 방사선육종 국화 품종 (아티다크초콜렛)의 티백 시제품을 제조
○ 저온 건조 방법 (온도 15 ℃, 습도 20%)을 시용하여 티백을 제조하고, 시판중인 노란색 국화차 티백을 구입하여 비교 분석함
나. 침출 온도와 시간에 따른 추출물 제조 및 항산화 활성 비교분석(논문 심사 중)
○ 티백 (1 g)을 75 ℃와 100 ℃의 물 (100 ml)에 각각 2분, 4분 동안 침출 후 동결건조함
○ 두 개의 다른 국화차에 대해 ABTS와 DPPH 라디칼 소거능 실험을 진행한 결과 아타다크초콜렛 차가 시판 중인 국화차보다 DPPH 라다칼 소거능이 높 았음
3. 양성자빔 활용 작물육종 기반기술 개발
3-1. 모델식물 활용 방사선원(감마선, 양성자)별 변이 메커니즘 규명 및 비교
가. 양성자빔 조사조건에 따른 모델식물 생체영향 및 감수성 데이터 구축
- 양성자빔 및 감마선 조사 M1 식물체 생체영향 분석
- 선원별‧선량별 애기장대 발아율, 생존율, 결실종자 무게 데이터 확보
- 방사선육종 관련 주요 선량 결정: LD50 (양성자빔: 1,051 Gy, 감마선: 1,087 Gy), 어깨선량 Dq (양성자빔: 787 Gy, 감마선: 900 Gy), Dq 50-75% (양성자빔: 394-590 Gy, 감마선: 450-675 Gy)
나. 고추 대상 양성자빔 생체영향 및 감수성 데이터 구축
- 방사선원별 LD50, Dq, Dq 60% 선량에 대해 각각 M2 400계통씩 육성
- 각 M2 집단에서 표현형 돌연변이 빈도 및 스펙트럼 분석: 양성자빔 조사 집단에서 총 16가지 변이 표현형, 감마선 조사 집단에서 총 9가지 변이 표현형 확인
다. 모델식물 생체영향 및 표현형 변이 상관관계 분석
- M1 세대 생존율을 기반으로 결정한 주요 선량 별 표현형 변이율 비교분석: 양성자빔의 경우 787 Gy(Dq 선량) 조사 집단(변이율: 5.25%), 감마선의 경우 1,200 Gy(LD50 근접 선량) 조사 집단(변이율 4.5%)에서 가장 높은 변이율이 확인되었음.
- 감마선 조사의 경우 엽색 변이가 엽형태 변이에 비해 7배 가량 높게 나타난 반면 양성자빔 조사의 경우 두 종류의 변이가 비슷한 비율로 나타나 유기 변이의 스펙트럼이 더 넣은 것으로 판단되었음.
라. 양성자빔 조사 변이체 전장 유전체 분석
- 양성자빔 주요 선량(LD50, Dq, Dq 60%), 감마선 Dq 선량에 대해 각 선량 별 4개체씩 전장 유전체 서열 분석: 원품종 서열의 평균 97.78% 영역을 포함하는 각 전장 유전체 서열을 확보함
3-2. 대표작물(14종)별 최적 변이 유기조건 확립 및 데이터베이스 구축
가. 대표작물 14종에 대한 양성자빔 MeV, dose 별 LD50, RD50 정보 구축
- 대표작물 14종 종자(배양체)의 감마선 및 양성자빔에 대한 LD50, RD50 정보 도출을 통한 양성자빔 데이터베이스 구축
- TR103 양성자가속기의 이온 가속에너지(MeV) 및 SOBP 기술 적용 여부에 따른 동양란의 생체반응 비교 결과 45MeV, SOBP 미적용 조건이 해당 분석 재료의 육종용 최적 조사조건으로 확인
- RFT30 20MeV(500nA) 양성자가속기의 육종용으로의 이용 가능성 평가 결과 사용이 제한적인 것으로 확인
3-3. 주요작물(2종) 양성자빔 생체영향 및 감수성 데이터 구축
가. 벼 대상 양성자빔 생체영향 및 감수성 데이터 구축
○ 벼 종자 대상 양성자빔 및 감마선 조사 후 생육반응 검정
- 100 MeV 양성자빔 및 감마선 조사 후 발아율, 생존율, 생육상태를 검정한 결과 동일 선량에서 감마선 대비 양성자빔이 주는 생물학적 영향이 큰 것을 확인하였으며 돌연변이 유기를 위한 적정선량으로 양성자빔은 100-150 Gy, 이에 상응하는 감마선량은 170-250 Gy를 도출
○ 벼 양성자빔 돌연변이 집단 구축을 위한 대규모 조사 및 초기생육 검정
- 우수 벼품종을 대상으로 돌연변이 집단을 구축하기 위해 적정선량을 적용 대규모 조사를 수행하였으며 초기생육 검정을 완료하고 본포장에 전개
나. 고추 대상 양성자빔 생체영향 및 감수성 데이터 구축
○ 벼 종자 대상 양성자빔 및 감마선 조사 후 생육반응 검정
- 100 MeV 양성자빔 및 감마선 조사 후 발아율, 생존율, 생육상태를 검정한 결과 동일 선량에서 감마선 대비 양성자빔이 주는 생물학적 영향이 큰 것을 확인하였으며 돌연변이 유기를 위한 적정선량으로 양성자빔은 300 Gy, 이에 상응하는 감마선량은 400 Gy임을 도출
