□ 연구의 목적 및 내용
본 과제에서는 비강투여를 통하여 내인성 Danger signal인 High Mobility Group Box 1 (HMGB1)의 억제 peptide 4종 및 HMGB1 siRNA와, 복합 기능을 가진 신경 보호단백질인 Osteopontin의 RGD motif를 포함하는 heptamer peptide를 비침습적으로 중추신경계에 전달한 후 뇌졸중 후 손상억제 및 보호 효과를 확인하고, 보완적 병용투여 조합을 선정하고자 함. 본 연구를 통하여, 비강 투여된 약물의 이동 경로, 전달 효율, 분비 kinetic
□ 연구의 목적 및 내용
본 과제에서는 비강투여를 통하여 내인성 Danger signal인 High Mobility Group Box 1 (HMGB1)의 억제 peptide 4종 및 HMGB1 siRNA와, 복합 기능을 가진 신경 보호단백질인 Osteopontin의 RGD motif를 포함하는 heptamer peptide를 비침습적으로 중추신경계에 전달한 후 뇌졸중 후 손상억제 및 보호 효과를 확인하고, 보완적 병용투여 조합을 선정하고자 함. 본 연구를 통하여, 비강 투여된 약물의 이동 경로, 전달 효율, 분비 kinetics 등을 규명하여 비침습적 비강투여를 중추신경계질환 치료를 위한 약물 투여의 현실적 대안으로 개발할 수 있는 실용적인 기반 정보와 근거를 제시하고자 함. 특히, 치료 약물의 비강을 통한 안정적 전달효율을 높이기 위하여 nano-carrier를 활용하여 비강투여 약물의 효율, 안정성을 높이고 분비 kinetics를 조절하고자 함.
□ 연구개발성과
○ 본 과제를 통하여 내인성 DAMP인 High Mobility Group Box 1 (HMGB1)의 HMGB1 A box 와 HBHP, Hpep1 등의 억제 peptide 4종 및 HMGB1 siRNA를 뇌졸중 동물 모델에 비강투여하여 뇌경색 억제, 항염증, 항산화, 2차 감염 억제등의 보호 효과를 얻을 수 있음을 확인함.
○ 복합 기능을 가진 신경 보호단백질인 Osteopontin의 RGD motif를 포함하는 icosamer peptide를 비침습적으로 중추신경계에 전달한 후 뇌졸중 후 손상억제 및 보호 효과를 확인하였고 펩타이드 크기를 RGD motif를 포함하는 7 amino acid까지 최소화하는데 성공하였으며, 상기펩타이드의 혈관 형성 유도, 포식작용 유도 등, 서서히 진행되는 뇌조직 회복/재건 효과를 확인함
○ Ethyl pyruvate 변이체 및 Neutrophil 활성화 (NETosis) 억제제 중 하나인 Cl-amidine와 같은 small molecule 의 비강투여 효과도 확인함
○ 상기 결과를 기반으로 acute한 신경 보호 효과와 서서히 진행되는 뇌조직 회복/재건 효과를 동시에 얻을 수 있는 보완적 약물의 병용투여 방안의 근거를 제시하며 가능한 조합을 제안함
○ 특히, 비강 투여시 nano-carrier 등을 활용한 치료 약물의 안정적, 지속적 전달을 통해 치료 효율을 현저히 높일 수 있음을 확인하였음
○ 본 연구를 통하여, 비강 투여된 약물의 치료 효과, 전달 효율, 분비 kinetics 등을 규명하여 비침습적 비강투여를 중추신경계질환 치료를 위한 약물 투여의 현실적 대안으로 개발할 수 있는 실용적인 기반 정보와 근거를 제시함.
□ 연구개발성과의 활용계획 (기대효과)
(1) 최적화, 최소화된 HMGB1 A box 유도체의 치료 펩타이드로의 개발을 위한 기본 정보 제공
(2) Osteopontin의 RGD motif를 포함하는 최적화, 최소화 peptide개발
(3) 약물의 보완적 병용 투여 개념 확립 및 최적 약물조합의 타 질환에의 적용

In this study, noninvasive administration of therapeutic peptides, including 4 inhibitory peptides of High Mobility Group Box 1 (HMGB1) and HMGB1 siRNA, which are endogenous dangerous signals, and RGD motif-containing peptide of Osteopontin, a neuroprotective protein. After the intranasal delivery to the central nervous system, we want to confirm the protective effect to stroke animal model and to select a complementary combination of the therapeutics.
The purpose of this study is to present basic information and evidence for developing non-invasive nasal administration as an alternative to treat central nervous system disease. In particular, we intend to use nano-carriers to increase the stable delivery efficiency through the nasal cavity of therapeutics.
□ Results
○ In this study, 4 types of inhibitory peptides such as HMGB1 A box and HBHP of High Mobility Group Box 1 (HMGB1), which are endogenous DAMPs and HMGB1 siRNA, were INTRANASALLY administered to the animal model of stroke and a robust protective effect such as suppression was confirmed
○ Non-invasive delivery of 7 aa peptide containing RGD motif of Osteopontin, a neuroprotective protein with multiple functions, confirmed a robust protective effect after stroke. The slowly progressing brain tissue recovery/ reconstruction effect, such as induction of blood vessel formation and phagocytosis was detected
○ We have also confirmed the protective effects of intranasally delivered small molecules such as ethyl pyruvate variants and Cl-amidine, one of theNeutrophil Activation (NETosis) inhibitors.
○ Based on the above results, we suggest the possible combinations of the complementary drugs that can simultaneously achieve acute neuroprotectiveeffect and slow progression of brain tissue recovery / reconstruction effect.
○ In particular, it was confirmed that the treatment efficiency can be significantly increased through stable and continuous delivery of therapeuticdrugs using nano-carriers, etc., during nasal administration.
○ Through this study, we provided basic information and evidence for developing non-invasive nasal administration as an alternative way to treat of central nervous system disease.
