□ 연구의 목적 및 내용
○ 최종목표: 줄기세포 미세환경 조절이 가능한 창상 치료용 생체모사 기반 고점착 패치 개발
○ 본 연구를 통해 개발된 생체모사 기반 줄기세포 창상 치료 효능 향상 고점착 패치는 1) 줄기세포의 환부 표면 부착성을 향상시키고, 2) 부착되는 줄기세포의 세포 생착율, 이식 후 생존율 증대, 치료용 성장인자 분비능 증진이 가능한 미세환경 조절도입이 가능하였음. 또한 문어 빨판 등에 기반한 생체모사 기법 도입을 통해 3) 비화학적 표면 고점착이 가능하여 창상 부위의 굴곡, 움직임 등 다이나믹 환경에도 점
□ 연구의 목적 및 내용
○ 최종목표: 줄기세포 미세환경 조절이 가능한 창상 치료용 생체모사 기반 고점착 패치 개발
○ 본 연구를 통해 개발된 생체모사 기반 줄기세포 창상 치료 효능 향상 고점착 패치는 1) 줄기세포의 환부 표면 부착성을 향상시키고, 2) 부착되는 줄기세포의 세포 생착율, 이식 후 생존율 증대, 치료용 성장인자 분비능 증진이 가능한 미세환경 조절도입이 가능하였음. 또한 문어 빨판 등에 기반한 생체모사 기법 도입을 통해 3) 비화학적 표면 고점착이 가능하여 창상 부위의 굴곡, 움직임 등 다이나믹 환경에도 점착이 잘 이뤄짐. 이러한 연구 결과를 바탕으로 창상 동물모델 도입을 통해 4) 창상 치료 효능 향상시키는 결과를 확보함.
□ 연구개발성과
○ 본 연구 개발의 결과인 줄기세포 미세환경 조절이 가능한 생체모사 기반 창상치료 패치시스템을 개발함.
- 다양한 굴곡과 움직임을 가지는 창상 표면에 화학적 접착성분을 사용하지 않고도 문어 빨판 등을 이용한 생체모사 기법 기반 안정적 고점착을 습윤도가 높고, 굴곡지며, 움직임이 있는 다이나믹한 환경의 피부에서 성공적 이뤄냄.
- 삼차원 구조체의 디자인을 기반으로 나노입자, 광생명조절기법 등의 새로운 줄기세포 자극 요인을 접목시켜 줄기세포 미세환경을 제어할 수 있는 기법을 확립함. 이를 통해 창상치료 효능이 증진된 줄기세포를 환부에 전달할 수 있었으며, 패치와 접목 시 간단하고 효과적인 시술 방식으로 활용될 수 있어, 전임상 실험을 통해 확인된 바와 같이 창상 치료효능을 증진시킴.
○ 본 연구팀은 지난 3년 간의 연구를 수행하여, Nature, Advanced Materials, Advanced Functional Materials, ACS Applied Materials and Interfaces 등의 유명 저널 포함 총 10편의 SCI(E)논문을 게재하였으며, 국내특허 등록1건, 국내 특허출원2의 성과를 달성함. 이는 초기 정량목표인 SCI(E) 논문 6편, 국내 특허출원1건, 국내 특허등록1건을 월등히 상회하는 목표치 초과달성 실적임.
□ 연구개발성과의 활용계획(기대효과)
○ 본 연구 개발의 결과인 줄기세포 미세환경 조절이 가능한 생체모사 기반 창상치료 패치시스템은 1) 다양한 굴곡과 움직임을 가지는 창상 표면에 화학적 접착성분을 사용하지 않고도 생체모사 기법을 통해 안정적으로 점착하며, 2)창상치료 효능이 증진된 줄기세포를 환부에 전달할 수 있기 때문에 간단하고 효과적인 시술 방식으로 활용될 수 있으며, 임상 실험을 거친 후, 즉각적인 상용화 및 제품화의 가능성이 매우 높음. 기존 창상 치료용 드레싱 제재들은 창상 환부에 단순히 부착되는 수준에 그치고 있으나, 본 연구에서 개발된 생체모사 기반 창상 패치는 내부에 줄기세포 부착, 배양 및 미세환경 조절이 가능한 삼차원 구조체를 적용할 수 있어 치료효능이 증진된 줄기세포를 접목시킬 수 있다는 새로운 개념의 창상치료 패치 기술이라 볼 수 있으며, 해당 원천기술 확보를 통해 줄기세포와 접목될 수 있는 치료용 생체모사 점착 패치라는 신산업 분야를 창출 및 선점할 수 있음.
○ 본 연구결과는 피부(창상) 외에도 다른 신체 내부 기관에 적용이 가능하여 다양한 의료분야에 응용될 수 있는 잠재력이 있으며, 국민 보건의료의 질을 높이는데 기여할 수 있음.

○ Research Contents: Developing technologies such as novel three-dimensional structure fabrication, nanoparticle technology, hypoxic environmental induction which can control cellular microenvironment. Also, developing patch with high-adhesion property for increasing surface adhesion effect onwound treatment with the stem cells.
○ The high-adhesion patch developed through this study to improve the efficacy of stem cell for wound treatment was developed.
1) Improving the surface adhesion of stem cells to the lesions. 2) Enhancing stem cell engraftment, cell viability, angiogenic paracrine factor secretion by controlling cellular microenvironment. Additionally, byadopting bio simulation techniques based on mimicking octopus structure, 3) non-chemical surface adhesion and adhesion to the dynamic environment such as bending and movement of the skin wound sites were aimed to be achieved.
Based on the previously mentioned techniques, we tried to 4) improve the efficacy of stem cells for wound healing.

○ We have developed a biomimetics based skin patch system that can control stem cell microenvironment for enhancing therapeutic efficacy fo the stemcell toward wound closing.

- In addition to the novel three-dimensional structure, bioreducible nanoparticles, photobiomodulation, and biodegradable polymers were incorporated to establish a technique to control the stem cellmicroenvironment. It was possible to deliver stem cells with improved woundhealing efficacy to the lesions.
- Existing wound treatment dressings only focused on improving attachment to the wound site. However, the biomimetic wound patch developed in this study can be additionally put three-dimensional structures or other technologies developed in this research that can control the stem cell microenvironment to improve therapeutic efficacy.
○ Our research team has conducted research over the last three years and published a total of 10 SCI(E) papers including famous journals such as Nature, Advanced Materials, Advanced Functional Materials, and ACS Applied Materials and Interfaces. Along with the achievement of 2 patent applications. This records exceed the quantitative goals which was originally suggested (6 SCI(E) papers, 1 domestic patent application, and 1domestic patent registration).
□ Expected Contribution
○ The result of this research can improve therapeutic efficacy of stem cells by controlling cellular microenvironmental for wound closing. Also, our result, novel patch system can be stably adhered to the wound surface with various bends and movements without using chemical adhesives. Since it is possible to deliver stem cells with enhanced therapeutic efficacy, wound treatment efficacy to the affected area easily, it can be considered as a simple and effective method for various future biomedical application.
After clinical trials, our result will gain a high possibility of immediate commercialization and commercialization. Existing wound treatment dressings are merely attached to the wound site, but the biomimetic wound patch developed in this study can be applied to skin surface with high adhesion property along with unique three-dimensional structure that can controlstem cell efficacy. It can be regarded as a wound treatment patch technology with a new concept of grafting stem cells with enhancedefficacy.
○ The results of this study can be applied to other internal organs besides skin (wounds), which has the potential to be applied to various medicalfields, and can contribute to improving the quality of public health care.
