연구의 목적 및 내용
1. 혈전증의 원인/위치/시기에 따라 Ly6C-hi아형 염증성단핵구와 Ly6C-lo아형 회복성단핵구가 혈전으로 이주하여 결합/축적되는 정도가 다를 것이라는 가설을 검증하고 기전을 규명할 것임
2. 단핵구 결핍 유발 및 knock-out생쥐 실험을 통해 동맥혈전에 결합된 Ly6C-hi아형 염증성단핵구와 Ly6C-lo아형 회복성 단핵구의 비율이 혈전 형성, 성장, 그리고 혈전용 해저항성에 영향을 줄 것이라는 가설을 검증하고 기전을 규명할 것임

연구의 목적 및 내용
1. 혈전증의 원인/위치/시기에 따라 Ly6C-hi아형 염증성단핵구와 Ly6C-lo아형 회복성단핵구가 혈전으로 이주하여 결합/축적되는 정도가 다를 것이라는 가설을 검증하고 기전을 규명할 것임
2. 단핵구 결핍 유발 및 knock-out생쥐 실험을 통해 동맥혈전에 결합된 Ly6C-hi아형 염증성단핵구와 Ly6C-lo아형 회복성 단핵구의 비율이 혈전 형성, 성장, 그리고 혈전용 해저항성에 영향을 줄 것이라는 가설을 검증하고 기전을 규명할 것임
3. Ly6C-hi아형 염증성단핵구와 Ly6C-lo아형 회복성 단핵구의 동맥혈전으로의 이주/결합/축적 및 이에 따른 혈전 특성 변화가 고지혈증/뇌경색 동반에 의해 교란될 것이라는 가설을 검증하고 기전을 규명함. 또한, 급성뇌경색 환자의 뇌혈관에서 제거된 혈전을 이용한 프로테옴 연구를 통해 단핵구 매개 혈전증 관련 염증성단핵구 행동제어 펩타이드-바이오이펙터를 발굴할 것임.
4. 상기 연구결과를 바탕으로 동맥혈전을 신속하게 영상탐지하고 동시에 염증성단핵구 행동제어를 통해 혈전성장을 억제하며 (MCP-1 inhibitor, CCR2 siRNA, 또는 발굴된 펩타이드-바이오이펙터 탑재), 또한 신생 혈전을 예방적으로 용해시키는 (thrombin에 의해 활성화되는 돌연변이 pro-urokinase 클로닝 및 탑재) ‘PrevenTheranostic 나노입자’를 개발할 것임
5. 상기 단핵구 행동제어 기반 예방-진단-치료 나노입자의 효능이 혈전증에 국한되지 않고 동맥경화-혈전증-뇌경색 스펙트럼을 포괄하는 고위험 뇌혈관질환 병태생리에 통합적으로 작용하도록 심화 발전시킨 ‘다기능 PrevenTheranostic 나노입자’를 개발할 것임. 이 과정에서 항염증 효과를 보이는 rapamycin 또는 염증성단핵구 감지-활성 광감작제의 추가탑재를 고려할 것임.

Based on mechanistic insights and novel peptide-bioeffctors derived from characterization studies to decipher the link between innate immunity and arterial thrombosis, we will develop an advanced class of theranostics (preventive therapeutics + diagnostics = PrevenTheranostics) that allows simultaneous imaging / prevention / treatment of monocyte-related atherothrombosis at multi-pathophysiologic milieus in a seamless manner.

2. Identified changes of leukocyte content of arterial thrombi over time
3. Demonstrated the leukocyte content of arterial thrombi between genders
4. Obtained the preliminary data that an increase of blood monocytes leads to a change in the monocyte and macrophage content in thrombi as well as thrombus size

Expected Contribution
1. The relationships between innate immunity and arterial thrombosis will be revealed and characterized for the first time and in detail.
2. As a new paradigm in nano-medicine, PrevenTheranostics will enable both imaging and preventive treatment of atherothrombosis and stroke in a timelier and more tailored way.
3. Inflammatory milieu-specific modulations of atherothrombosis pathophysiology and prophylactic thrombolysis will improve the limited risk-benefit profile of current anti-thrombotic therapy, which heavily depends on non-specific inhibition of the thrombosis / coagulation pathway and thus suffers frequent bleeding complications.
