전 세계적으로 신종마약류(New psychoactive substances, NPS)의 남용은 심각한 사회적 문제로 대두되고 있으며, 다양한 NPS가 인간에게 많은 악영향을 미치고 있다. 최근 NPS 출현이 급증하면서 많은 남용사례가 보고되고 있으며, NPS의 임시마약류로 지정도 급격히 늘어나고 있다. 본 과제는 3년 동안 3C-E 등 15종 이상의 임시마약류에 대하여 중추신경계 작용, 일반약리, 대사체 연구 등을 수행하였다. 중추신경계 작용 평가를 위해 랫드 선조체로 부터 시냅토좀을 분리하여 약물에 의한 시냅토좀의 도파민 변화량을 LC-MS/MS로 분석하였고, in vivo 미세투석법을 이용하여 시험물질에 의한 도파민 농도 변화를 분석하였다. 약물의 약리학적 특성을 파악하기 위해 시험물질의 단회투여 후 일반적인 운동활성을 관찰하였으며, 사람의 간 마이크로솜을 이용하여 대사체 분석을 하였다. 또한, 성별에 따른 동물 행동약리학적 의존성 차이를 평가하기 위하여, Diazepam 등 마약류 8종에 대한 조건장소선호도, 자가투여시험을 수행하였다. 본 과제의 연구결과, 시냅토좀에서의 도파민 변화는 15종 물질 중 5종에서 농도의존적으로 또는 일부 농도에서 증가하였다, 미세투석법에서는 8종의 임시마약류에서 도파민 농도가 증가하였다. 일반운동활성시험에서는 15종 중 5종에서 운동활성이 유의적으로 증가하였다. 사람 간 마이크로솜을 이용한 대사체 분석 결과, 6종 물질에서 5∼13가지의 대사체가 검출되어 구조를 예측하였다. 성별에 따른 의존성평가에서는 마약류 5종에서 암컷이 더 민감하게 의존성을 나타냈다. 본 과제의 연구결과는 임시마약류의 마약류 지정 시 과학적 근거자료가 되며, 임시마약류를 포함한 NPS의 의존성 등 유해성 평가 시 기초자료로 활용될 것이다.

The problem of new psychoactive substances (NPS) abuse is emerging globally, and various NPS have caused adverse effects in human. With the recent surge in the emergence of NPS, many cases of drug abuse have been reported, and the number of substances designated as temporary drugs has also increased rapidly. This study was performed to evaluate effects on the central nerve system, the pharmacological changes and the metabolic pathway with more than 15 temporary narcotics, including 3C-N, throughout 3 years. The effect of central nervous system was examined with synaptosomes isolated from the rat striatum, and the concentration of dopamine in synaptosomes was analyzed by LC-MS/MS. The change of dopamine concentration caused by the test substance was evaluated using microdialysis in the rat striatum. In order to identify pharmacological characteristics of the drugs, the locomotor activity of mice was observed after singly administered, and their metabolites using human liver microsomes were analyzed by LC-MS/MS. In addition, to study the gender impact of the dependency assessment, 8 narcotics, including diazepam were evaluated through behavioral pharmacological test, such as conditioned place preference test and self-administration test. From the results, dopamine concentrations in synaptosomes increased in dose-dependent manner or in certain concentrations treated with five temporary drugs. Eight temporary narcotics led to increase of the dopamine concentration in microdialysis test. In the locomotor activity test, 5 out of 15 temporary narcotics showed a significant increase. The metabolites from 5 to 13 were detected in six temporary drugs and their structures were presumed. In dependency evaluation according to gender, female rats showed more sensitive dependent behavior when treated with 5 narcotics. Consequently, these results through this project might be used as scientific evidence when temporary narcotics are designated to narcotics, and also as preliminary data for dependency assessment of NPS, including temporary narcotics still not evaluated.
