최종 목표
최근에 연구되는 뼈조직 재생기술은 신체의 재생 능력을 최대한 활용하기 위하여 손상되 조직에서의 줄기세포의 이동, 성장 및 분화 뿐만 아니라 항염증 및 혈관형성을 촉진시키는 지지체를 제작하는 것을 중점으로 한다.
산화 세륨 나노입자(CNP)는 항산화 특성을 기반으로 염증을 감소시키고, 혈관생성을 촉진시키는 등 뼈조직 재생용 소재로 매우 매력적인 특징을 가지고 있다. 그러므로 본 연구에서는 PCL나노지지체에 CNP를 코팅하여(CNP@PCL) 뼈조직재생을 촉진시키는 새로운 시도를 하고자

산화 세륨 나노입자(CNP)는 항산화 특성을 기반으로 염증을 감소시키고, 혈관생성을 촉진시키는 등 뼈조직 재생용 소재로 매우 매력적인 특징을 가지고 있다. 그러므로 본 연구에서는 PCL나노지지체에 CNP를 코팅하여(CNP@PCL) 뼈조직재생을 촉진시키는 새로운 시도를 하고자 한다. 이러한 전략은 CNP의 항산화, 항염증 및 혈관생성 촉진능과 PCL의 우수한 생체적학성 및 기계적 특성을 모두 활용할 수 있을 뿐만 아니라, 나노지지체 구조와 그 표면에 코팅된 CNP입자의 나노표면의 통해 줄기세포의 부착, 증식 및 분화를 향상시킬 수 있기 때문이다.
이번 연구에서 최적화된 CNP@PCL 나노지지체는 항산화, 항염증, 혈관형성 능력을 통한 뼈 조직 재생을 극대화 시킬 수 있는 플랫폼을 제공하여 다양한 모양의 뼈조직 지지체 개발을 위한 기반연구가 될 수 있을 것이다.

1. Fabrication of CNP@PCL scaffolds
-Synthesis of PCL nanofibers by electrospinning method.

-Degradation in PBS and apatite forming ability in SBF with different time points.
-Evolution of Physico-chemical properties (XRD, SEM, TEM, TGA, BET, XPS, FTIR, and Raman)
-Evolution of surface roughness (AFM), mechanical properties (Instron), stiffness (Nanoindentation), and wettability (contact angle) Fabrication of CNP@PCL scaffolds and Physico-chemical properties will be confirmed.
2. CNP@PCL scaffolds in in-vitro environment with three types of cells (hMSC, Raw264.7, and HUVEC)
-hMSc cell with scaffolds: Cell proliferation rate, cell adhesion, cell attachment, and cell morphology characterized by markers staining and confocal images
-Osteogenic differentiation by q-PCR
-In vitro studies with RAW264.7 cell: ROS and macrophage response by q-PCR
-In vitro studies with HUVEC cell: cell adhesion, tube formation, angiogenic response by q-PCR
-Mechanism pathway by q-PCR and western blot l Cell-materials infraction, differentiation, pathway will be confirmed.
3. In vivo animals model for bone regeneration
-In-vivo studies: Scaffolds will be implanted for two weeks and four weeks.
-H & E stained will be observed under microscope to determine in vivo biocompatibility and angiogenesis.
-Bone formation will be performed by Micro-CT image.
-Angiogenesis assay using the chick chorioalantoic membrane (CAM) model
□ Bone regeneration will be confirmed. This material will be used for tissue regeneration.
연구개발성과
- The results can confirm that promoting vascularized osteogenesis in the interior parts of the implanted scaffold can minimize the high clinical failure rates observed with natural and synthetic bone grafts and thus initiate further research and development toward clinically relevant bone scaffolds.
- The results can lead to the introduction of innovative strategies and methodologies that can be further developed in academia and biomedical sectors to innovate further ideas to overcome the current drawbacks of the traditional tissue engineering scaffolds.
- Biomedical fields related to bone regeneration, such as bone deformities, bone loss, osteoporosis and other bone regeneration therapy, orthopedic, oral and maxillofacial surgery and other bone diseases can also benefit from developing such novel biomaterials.

- Shedding light on the technical limitations in conventional tissue engineering scaffolds that are often designed without appropriate consideration for the translational potential of the scaffold to clinical applications.

(B) Economical and industrial aspects
- The technology of fabrication of the biomaterial coated scaffolds introduced in this project is cost-effective and can be scaled-up for large industrial-scale production of bone tissue engineering scaffolds without major problems. So, it corresponds to a new technology of future value for the company's growth and development. Also, it can attract domestic and global companies' investments. Furthermore, it is expected to keep the national and international standards and further develop them.
