Vascular endothelial growth factor (VEGF) is known as a potent mediator of brain tumor angiogenesis, vascular permeablity, and glioma growth. Using immunohistochemisty, the relationship between the expression of VEGF and microvessel count and density were evaluated in 62 surgical cases of glioma including 33 low grade astrocytoma, 6 anaplastic astrocytoma, and 23 glioblastoma. In order to determine the apoptotic index (AI), in situ end-labeling method was applied in gliomas of different grade of malignancy. Immunostaining for VEGF were observed in the tumor cell cytoplasm. Of all cases of glioma, 44 tumors (77%) were weak to strong postive for VEGF and 18 cases exhibited no tumor cell immunoreactivity. VEGF positivity was correlated with grade of malignancy in gliomas; 22 glioblastoma (96%), 5 anaplastic astrocytoma (83%), and 17 low grade astrocytoma (52%). Mean microvessel count and density were significantly higher in VEGF-positive tumors (75.7 and 4.1 %) than in VEGF-negative tumors (43.9 and 2.5%). Apoptotic cells were readily found in all the examined tumors; in most cases the AI values were below 5%. The highest value of AI was observed in glioblastoma (8.6%) and the lowest in an anaplastic astrocytoma (1.9%). It seems that the grade of malignancy was not related with AI values. These results suggest that VEGF play a role in the growth and progressive angiogenesis of the human glioma.
