Morphological and ultrastructural characteristics of the hemocytes, the hemopoietic organ, the pathway of hemocytic differentiation and the cellular immune responses from the grasshopper, Euprepocnemis shirakii Bolivar were described using phase contrast microscope, scanning and transmission electron microscope. The hemocytes were of six types: prohemocytes, plasmatocytes, granulocytes I, granulocytes II, spherulocytes and oenocytoids. The prohemocytes were small and spherical cell with large nucleus. The plasmatocytes were spindle-shaped or polymorphic cells which shown numerous cytoplasmic processes on the cell surface and they had lysosomes and vesicles that may be invloved in phagocytic function. Especially, multivesicular bodies were observed in the polymorphic cells. The granulocytes I were spherical-shaped cells. They were characterized by a number of electron dense granules measuring 0.2-0.3 ㎛ in average diameter and marginal bundles of microtubules which were always in close proximity to the cell membrane. The granulocytes II were oval or spindle-shaped cells. They contained large electron dense granules measuring 0.5-0.8 ㎛ in average diameter. Their cytoplasm was filled with numerous granules. The hemopoietic organs located on the upper surface of the alary muscle between 1st and 8th segments, and generally consisted of 3 kinds of cells: the reticular cells, stem cells and differentiating hemocytes. The reticular cells had numerous cytoplasmic processes and formed a complex network. Characteristically, pinocytotic vesicles and rough endoplasmic reticulum were well developed in this cell. The stem cells gave rise to differentiating hemocytes of the different cell lineages: prohemocytes, plasmatocytes, granulocytes I, granulocytes II and spherulocytes. Hemocytic immune responses to the injection of foreign materials such as colloidal gold solution containing gold particles with average diameter of 10 nm into the hemocoel of grasshopper, Euprepocnemis shirakii Bolivar were examined using electron microscope. Only the plasmatocytes which consist of about 28% of total hemocytes participated in this response by the form of phagocytosis, but the other kinds of hemocytes were not reacted. The initial phagocytic response of plasmatocytes was composed of both the first projection of cytoplasmic processes and the second formation of phagocytic vesicles by the surface reaction of the plasma membrane. And this procedure was finished within 10 min after injection. The phagocytic vesicles within the plasmatocytes were comprised electron lucent fibrous or membranous inner structure initially, changed to electron dense granular form after fusion of the primary lysosome, and then developed to secondary lysosome composed of multivesicular body in the next stage. The injection of bacterial suspension(5x10^6 cell/ml) led to the rapid formation of phagocytosis in plasmatocytes, granulocytes I and granulocytes II within 10 min, but nodule formation did not occure. Phagocytosis was accompanied by an initial reaction of forming numerous cytoplasmic process on the cell surface. After recognition of the hemocytes to the bacteria, cytoplasmic processes surrounded the bacteria and fused with the plasma membrane within 10 min after inoculation. As they formed phagocytic vesicle which moved to the interior of the cell. Active primary lysosome accumulated around the phagocytic vesicle and fused with limiting membrane of phagocytic vesicle, then developed to secondary lysosome within 1 hr. Digested bacterial residues were finally released from the cell by exocytosis within 9 hr after injection. In hemocytic encapsulation response after implantation of the spun polyester, multilayered capsule around the spun was formed within 6 hr after injection and three types of hemocytes - granulocytes I, granulocytes II and plasmatocytes - were involved in this response. The initial response by the blood cells was the lysis of granulocytes I on the surface of the implant. Next the granulocytes II attracted to the reaction sites envelope the implants, and form a multilayered capsule composed of three morphologically distinct regions. The inner layer of the capsule attached on the surface of the spun was composed of lysed granulocytes I and compressed granulocytes II. The middle layer of the capsule was comprised of three types of flattened hemocytes composed of granulocytes I, granulocytes II and plasmatocytes. Adjacent plasma membranes of these hemocytes were attached by desmosome-like structure, intermediate junctions and gap junctions. The outer layer of the capsule was composed of 1-2 layers of granulocytes II identical to normal free hemocytes found in the circulation, and along the boundary region to hemolymph 35 nm thick "outer sheath" representing termination of the capsule was identified. The injection of live Escherichia coli(ATCC29622) into the hemocoel of adults of grasshopper, Euprepocnemis shirakii, resulted in a phagocytosis of the number of reticular cells and differentiating hemocytes in the hemopoietic organ. Injected bacteria were rapidly phagocytosed by reticular cells 10 min after the injection. The phagocytic vesicles interacted with the primary lysosome so that a range of enzymes discharged onto the ingested bacteria. The digested bacteria in phagocytic vesicles were changed to electron dense debris. The reticular cells had considerable phagocytic activity to engulfe a large number of bacteria. As a results, many reticular cells undergone lysis, whereas others have engulfed fewer bacteria, succeeded in breaking down the bacteria.
