The optimum condition for the growth of recombinant Saccharomyces cerevisiae was determined as follows : temperature; 30℃, pH: 5.0, D.O. ; greater than 25% (v/v), and agitation speed; around 500 rpm. Casamino acid, as a nitrogen source, showed a tendency which enhance the cell growth within a range of concentrations examined(0.55 to 2.2% (w/v)). Plasmid stability of RS 20B-12 was relatively high through 30 generations. In the batch culture of recombinant Saccharomyces cerevisiae 20B-12 strain, the glucose concentration was increased gradually from 5 to 20%, the maximum dry cell weight and volumetric productivity(P_(v)) was also increased from 8.9 to 11g/l, and from 0.25 to 0.31 g/l/hr, respectively. However, the growth yield on glucose(Y_(s)) was decreased from 0.14 to 0.055, while the maximum ethanol concentration was increased 4 times from 20 to 80 g/l. These results were showed that the substrate inhibition had been triggered when glucose concentration was higher than 5%, either in the batch culture or in the flask culture. Therefore, in order to overcome substrate-level inhibitory effects, fed-batch culture technique with constant feeding rate was chosen as an experiment system for HBsAg production.
