A study was performed on the effect of Al ion aftertreatment on diffusion within Ca-alginate gel beads for cell immobilization carrier. As alginic acid is a kind of the polysaccharide extracted from the brown seaweeds, sodium alginate is purified alginic acid with a sodium salt and this becomes to gelation with poly metal-ion(Ca, Cu, Zn, Al etc.) by ion exchange. Sodium alginate 1.5%(W/V) solution was obtained as drops, and was later made to beads contacted in 50mM and 100mM CaCl(2) solution for 30~60 minutes, respectively. Although the pore size of alginate beads was of about 100Å order and its pore structure was suitable for cell immobilization, there was such a problem as the pore structure was destroyed by phosphate anion in culture medium. To bring this problem to settlement, the beads were aftertreated with 50mM Al(NO(3))(3) solution for 30 minutes and the kinetic diffusion study was done measuring the transient concentration of aqueous L-tryptophan solution contacted with one thousand beads with and without Al ion aftertreatment, respectively. It was found from the experimental result that these experimental data were in good agreement with the bulk diffusion mechanism of L-tryptophan in the pure water system. Since the Al ion aftertreatment made the alginate beads stabilized against phosphate anion and did not give rise to pore diffusion effect within the alginate beads, the pore structure of Al-treated alginate beads may be improved for cell entrapment. Therefore, it can be seen that the Al-treated alginate beads was suitable for cell immobilization in the view point of physical properties, chemical stability and pore diffusion.
