In this study, we made a target-sensitive liposome including monoclonal antibody and characterize their destabilization effects. Our newly developed technique for the coupling of monoclonal antibodies to liposome was used to study the specific delivery of Doxorubicin (DOX) and Methotrexate (MTX) to cellular targets in vitro . PE-liposomes containing Calcein, DOX and MTX were prepared by palmitoyl-antibody against determinants of molecules encoded by the major histocompatibility complex of the murine H-2K^b haplotype. DOPA(dioleoyl phosphatidic acid) was also added to stabilize PE-liposome and to increase drug encapsulation rate. These were incubated with various types of target cells, only H-2K^btypes of target cells(EL-4, RMA)showed evidence of Calcein releasing and drug effect, demonstrating that interaction of the liposomes with target cells was specific. However the therapeutic potential of this system will be realized after concerns about RES avoidance, stability in circulation, until target binding, and cellular uptake of released drug before diffusion away from the cells in vivo are addressed.
