This study was performed to investigate the inhibitory effects of calcium channel blocker, diltiazem, on the oxidative damage of lipids and proteins in free radical reaction induced by iron-dextran. The Sprague-Dawley strain male rats were devided into four groups: normal group(control), iron group, diltiazem injection group(DIL), and vitamin E injection group(VTE). The control group was treated with physiological saline solution. DIL group and VTE group were pretreated with intraperitoneally 24mg of herben and 50mg of dl-α-tocophorol per kg of body weight; and three groups, except normal group, were injected intraperitoneally 500mg iron per kg of body weight. The injection was carried out one single dose at 3hours after the last treatment. The injection was carried out one single dose at 3hours after the last treatment with the agents. We have evaluated the effect of diltiazem and iron as well as in combination on blood biochemical change, histopathological examination. Also we assayed for accumulation of hepatic lipid peroxidation and protein oxidation by-products by MDA, carbonyl compound, erythrocytolysis and for antioxident defense mechanism by antioxidant enzyme and glutathione redox cycling system in liver tissue, cytosol, microsome and mitochondria. The diltiazem protected decrease of CYP450 and inhibited the production of MDA andcarbonyl compound, the level of calcium and phosphatase in liver as compared with those of irontreated rats. The activities of SOD, CAT and GPx was increased. We speculate that the oxygen radical scavenging activities of the diltiazem might play a key role in the mechanism opposing the progression of iron treated hepatitis, but the activities of GSH and GST were decreased. The results suggest that calcium channel blockers provide antiperoxidative protection to liver and that the mechanism mainly due to the protection of CYP450 contents, the role as CCB, and the increase of SOD and CAT activities rather than glutathione metabolizing system. The administration of diltiazem protects against iron-induced hepatic toxicity in vivo. The protective effects seem to be caused by the prominent antioxidant activity of this compound.
