Among four fractions, ethylacetate(E.A) layer showed the strongest antioxidative activity in DPPH(1,1-diphenyl-2-picry hydrazyl) method. Yield of extracted that Vitis coignetae were 0.732g(4.1%)[he- xane], 3.0262g(16.56%)[Ethyl acetate], 5.71g(31.248%)[n-butyl alc- ohol], 8.028g(43.9%)[Water] and Vitis vinifera L. were 1.342g(2.2%)[hexane],15.72g(25.9%)[Ethyl acetate], 29.25g(48.16%) [n-Butyl alcohol],12.265g(20.19%)[Water]. Among the fractions, E.A layers showed a strong antioxidant activity than other layers in DPPH method and was similar to quercetin, which was a strong antioxidant. This fractions scavenged 83.4%, 84.6% of free radicals. The E.A layer were chromatographed on sephadex LH-20 and fractionated to seven fractions(Vitis coignetae → gⅠ∼gⅦ, Vitis vinifera L. → GⅠ∼GⅦ). After passing column, we gain two fractions(gⅤand GⅥ) that showed a very strong antioxidative effect in TBA(2-tiobarbituric acid), microsomal lipid peroxidation, PC(phosphatidyl choline)-liposome peroxidation and FI-CL. And this fraction was confirmation by TLC(Thin layer Chromatigraphy). In TBA mothod, we used three initiators. G-Ⅴ and G-Ⅶ inhibited strongly AlNH4(SO4)2/ascorbic acid-induced MDA generation, its inhibition activity was 10% higher than Quercetin. Also, In enzymatic(Fe3+-ADP/NADPH-I- nduced lipid peroxidation)systems, non-enzymatic(Fe2+/ascorbic acid-induced lipid peroxidation)system, gⅤ, GⅦ showed strong antioxidative effects against control. Compare with PC-liposomal peroxidation was intiated by the addition of AAPH(2,2'-azobis(2-amidinopropane)hydrochloride), g- Ⅴ, GⅥ and quercetin represented strong antixidant activity effect. The results suggest that Vitis coignetae, Vitis vinifera L. Have very strong antioxidant activity, inhibiting formation PC-OOH and MDA, in vitro.
