Currently used method for the measurement of capillary blood glucose is enzymatic assays using invasive type equipments. It requires patients to prick their fingers to take small blood samples several times a day. Then many diabetic patients avoid frequent and continuous blood glucose monitoring and involve some risk of the complications such as eye, kidney, and nerve disorders. To solve the problems of invasive method, a noninvasive method for blood glucose measurement using near infrared spectroscopy was developed. A portable near infrared (NIR) system which was newly integrated by our lab has been used to develop a non-invasive blood glucose monitoring. The portable NIR system includes a tungsten halogen lamp, a photo diode array type-InGaAs detector, and specialized reflectance fiber optic probes. Calibration models are generated for glucose in buffer solution over the 0500 mg/dL concentration range by use of partial least squares regression (PLSR) analysis of near infrared spectra from 1100 to 1750 nm. The resulting calibration models provide standard error of prediction (SEP) of 19 mg/dL. NIR reflectance spectra of different parts of human body (finger tip, earlobe, and inner lip) were acquired using specialized reflectance fiber optic probes. The shape of probes is composed of two parts, one for illumination into sample and the other for receiving the radiation from sample. Three kinds of probes with different distance between illumination and receiving part, such as 0.03, 0.1, and 0.5 mm, were investigated for optimization. The spectra were collected over the spectral range 1100 --1730 nm. PLSR was applied for the calibration and validation for the determination of blood glucose levels. Calibration modeling results were compared based on the kinds of probes and the measured human body parts. The results of calibration models showed a specific trend by the distance of fiber optic probe. The best result for ear model is SEP of 33 mg/dL with 0.03 mm distance probe. The best result for inner lip model is SEP of 33 mg/dL with 0.1 mm distance probe. The best result for finger tip model is SEP of 25 mg/dL with 0.5 mm distance probe. This trend indicated that the distance of fiber optic probe had an effect on penetration depth into skin tissue of human body and the optimum distance of fiber optic probe according to the parts of human body should be considered. This study provides the useful information concerning sample presentation for non-invasive blood glucose monitoring and demonstrates the feasibility of using a portable NIR spectrometer for the noninvasive measurement of blood glucose.
