This study investigates the differences in HSP70 expression by western blotting and immunohistochemistry in three groups:a no bath (control) group, hot bath only group, and a hot followed by cold bath group. Materials & Methods:Twenty seven white rats (200-230g) were randomly divided into three groups:a control group, a hot bath only group, and a hot followed by cold bath group. The hot bath consisted of immersion in a 41-43℃ water bath for 10 min, and the cold bath consisted of immersion in a 18-20℃ water bath for 3 min. Each bath protocol was performed twice. Bathing was performed three times a week for a total of four weeks. At the end of four weeks, rats from each group were randomly selected and sacrificed. Testes were obtained from each group. Results:Western blotting analysis of HSP 70 significantly expressed in three groups. and was more increased 1.5 times expression in the hot bath group than that of the no bath group. Similarly, Immunohistochemical analysis of HSP70 expression demonstrated significant elevations in the hot bath only group and HSP 70 immunoreactivity was found in Leydig cell and fibroblst in three groups and the levels of HSP70 expression in the control and the hot followed by cold bath groups were similar. Conclusions:The results of this study demonstrate elevations in HSP70 expression in the hot bath only group, which suggests induction of a coping mechanism for exposure to high temperatures. And, as the levels of HSP70 expression in the control and hot followed by cold bath groups are comparable;it appears that the levels of HSP70 are consistent with those seen in normal spermatogenesis.
