IM SUN 31002는 laccaes 활성도가 가장 높게 나타났다. 다른 균종에서도 laccase의 생산은 확인할 수 있었으나, 그 활성도는 높지 않았다. laccase의 활성도는 상수리나무 목분을 첨가한 배지에서 더 높게 나타났으며, COP 20242는 xylidine 첨가에 의한 laccase 활성도의 증가가 확인되었으나, 다른 균종의 경우 이 같은 효과는 나타나지 않았다. 부후 목재의 미시형태적 특징을 관찰한 결과 상수리나무가 소나무 보다 부후가 더 많이 진행되었다. 소나무와 상수리나무 모두 백색부후의 전형적인 특징인 세포벽의 침식과 박벽화 등이 관찰되었다. 특히 COP 20242는 중간층만 남겨두고 2차벽의 대부분이 분해되어 전형적인 백색부후균의 부후형태를 나타내었다. Therefore, this study was conducted to investigate the production of ligninolytic enzymes by, and decay characteristics lignin degradation pattern of the brown-rot fungus, Coniophora puteana which is a typical brown-rot fungus often used by many researchers as a representative test organism. In addition, an attempt was made in this study to see if four different fungal strains of C. puteana have the same behavior in degrading hardwood species. All four C. puteana strains showed positive reaction to Bavendamm reaction, but had different levels of laccase activity, which was the highest for the strain IM SUN 31002. The production of laccase was also detected for other strains but its activity was relatively low. Higher laccase activity was detected with the culture medium mixed with hardwood flour from Oak (Quercus acutissima C_(ARRUTH)), and with softwood flour from pine (Pinus densiflora S_(IEB) et Z_(UCC)). The laccase activity of strain COP 20242 increased when xylidine was added. However, other strains also showed this effect with addition of xylidine. The observation of micromorphological characteristics showed that Oak wood was more severly degraded than pine wood. Furthermore, cell wall erosion and thinning, typical degradation pattern of white-rot fungi, was caused by both species. Particularly, a strain (COP 20242) degraded most of the secondary wall except middle lamella, showing a typical decay characteristics of white-rot fungi. The laccase activity of all strains of C. puteana was consistent with the Bavendamm reaction result, but was not in agreement with the weight loss percentage of decayed wood and micromorphological decay characteristics. The results of this study showed thar C. puteana produces laccase, and that lignin in cell walls is degraded by laccase, which C. puteana produces as well as by other enzymes. The results of this study also suggest that the degradation mechanism by the ligninolytic enzyme systems needs to be reviewed.
