This study was carried out to investigate the content of isoflavones and estrogenic activity in Puerariae Radix. And HPLC using a dual column system was used to determine the content of isoflavones in Puerariae Radix. The simultaneous analytical time of isoflavones(daidzin, genistin, daidzein, glycitein, genistein) required less than 25 min per sample. It could shorten pretreatment time that extracted in 80% methanol and cut off disturbance peak detected before isoflavones peak. The content of daidzin(5,026.6 ~ 18,547.8 ,㎍/g), genistin(488.3 - 1,201.8 ㎍/g), daidzein(316.8 - 876.8 ㎍/g), genistein(8.4 ~ 63.1 ㎍/g), puerarin(l.6 - 10.84 g/100 g) in Pueraria thunbergiana showed higher levels compared to content of daidzin(l68.2 ,㎍/g), genistin(62.5 ㎍/g), daidzein(46.2 ,㎍/g), puerarin(0.03 g/l00 g) in Pueraria mirifca. Glycitein was not detected in Puerariae Radix. In uterotrophic assay for indicator of estrogenic activity, the Pueraria mirifica showed strong estrogenic activity but Pueraria thunbergiana and Butea superba did not show not estrogenic activity. Therefore, the content of isoflavones in Puerariae Radix could not be responsible for estrogenic activity.
