The absence of three-dimensional structure for CAs has been a handicap for progress of protein engineering, but the structure of a class I CA from Pesudomonas diminuta(CAD) was recently determined at 2.0Å resolution. In the report, an attempt is carried out such that the active-site residues of CAD were mutagenized to introduce higher deacylation activity with regard to CPC. The design of mutations was guided by the ligand bound structures of CAD the residues. The mutations are focused on the active-site residues, which are interacting with substrate(GL-7-ACA), especially for residues forming the side-chain pocket for the purpose of generating a sterically favored conformation to accommodate the α-aminoadipyl moiety of CPC.
In order to confirm the effect of the ten residues in active-site on catalytic activity, the residues involved in substrate binding were mutagenized to different amino acids and their effects for catalysis with regard to GL-7-ACA and CPC were investigated by comparing the deacylation activity of mutant CADs with wild-type. Three mutants, such as Y33βS and Q50βM, improved deacylation activity to CPC 120 and 180% relative to wild type. Since the Q50βM provided a starting point to improve deacylation activity to CPC, we take the mutant to add subsequently site-directed mutation to further improve deacylation activity. In rotating the torsional angles of the modeled α-aminoadipyl moiety, it collides with neighboring residues such as Y33β, 149α and F58β in a chosen orientation. In order to generate space for the α-aminoadipyl moiety, the first set(Y33β, 149α, F58β) was mutagenized to any amino acids by saturation mutagenesis. All mutants except for Y33βS and Y33βG showed higher deacylation to CPC than wide-type. Surprisingly, Y149αK and Y33βD showed greatest improvement in deacylation activity with regard to CPC as much as 600% and 450% of wide-type, respectively
