According to the previous reports, when aggregated AP1-42 was immunized into 7-month-old of Tg2576 mouse, which contains overexpressed human Swedish APP mutant gene and starts expressing AP deposition at the age of 8 month, A(3 deposition was effectively reduced. Secondly, previous data from AD patients reported that control subject showed relatively high concentrations of anti-AP antibodies in serum compared to AD patients. In this study, potential prevention and diagnosis for AD using anti-AP antibodies are discussed. To specify the therapeutic target for AP clearance in the animal model, aggregate forms of A(31-42 were injected into young (4-month-old) and adult (8-month-old) Tg2576 mice for 12 weeks. First of all, those groups showed the increased levels of anti-A(3 antibodies in serum, indicating that 12-weekimmunization produced the immune response in their bodies. Even after 12-weekimmunization, the high levels of serum titers lasted at the age of 11 months in two groups. Mice were sacrificed at 13 months then A(3 levels in brain were measured. Both A(340 and A4342 levels of brain decreased in immunized group compared to those of control. Furthermore, immunized group showed the decreased number of congo red stained amyloid plaques in the brain.
