1970년 Laemmli에 의해 확립된 전기영동 방법은 단백질의 순도 결정, 단백질의 구조적 변화에 관한 연구등을 위한 단백질 정제에 응용되고 있다. 이러한 전기영동 방법의 광범위한 응용을 위해서는 겔상에 분리된 단백질을 확인하는 과정이 필수적이다. 지금까지 보고된 단백질 확인 방법은 CB staining, silver staining, fluorescent staining, specific enzyme visualization, 그리고 autoradiography, scintillation spectrometry를 이용한 radioactive detection method가 있다. 이 방법들 중에서 CB staining은 가장 널리 쓰이고 있는 detection method이지만 감도가 silver staining 등에 비해 낮고 염/탈색에 소요되는 시간이 길다는 단점이 있다. 따라서 CB의 단점인 감도 및 소요 시간적인면을 개선하고자 본 실험을 행하였다. 본 연구에서는 ion pairing 기전을 이용하면 겔 상의 단백질 염색이 용이하다는 예비실험 결과에 따라 수종의 염색보조제를 검색하였으며, 그 중 Bismark Brown-R(BBR)을 이용하여 CB 염색의 감도 및 소요시간을 개선하였다. 염색 보조제 BBR의 혼합비를 결정하기 위하여 0.2% CB용액에 가해질 0.05% BBR 용액의 적정 혼합비를 조사한 결과 CB 1에 대하여 BBR 0.75의 혼합비에서 가장 효과적인 염색결과를 얻었다. 염색 용액의 pH는 2.8에서 가장 높은 염색 효과를 나타냈으며 pH 7.8의 alkaline 에서는 그 효과가 급격히 감소하였다. 상기 조건을 이용하여 즉 CB(0.2%):BBR(0.05%) = 1:0.75 비율로 methanol:acetic acid:water = 40:7:53의 용액에서 단백질을 염/탈색하였을 때, 이를 수행하는데 걸리는 시간은 20/25분 이었으며 BSA를 0.01-5 ㎍ 범위 내에서 염색한 결과 25 ng의 BSA까지 검지 가능하였다.

Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) has become one of the most powerful methods for separating proteins and determining their molecular weights. Various procedures based on the staining of proteins in polyacrylamide gels with different dyes [Coomassie blue, silver, amido black, etc.] have been extensively described. Among them, CB staining is the most widely used method. But it requires long times for staining/destaining of gels and gives the high background which makes sensitivity rather low as much as 100 ng of proteins. Silver staining has been considered as the most sensitive non-radioactive method and thus can be used to detect femtograms of proteins. However, this method has the drawbacks arising from high background, laborious multiple steps and toxicity of formaldehyde. Fluorescent reagents such as fluorescamine and ο-phthalaldehyde have also been used for visualizing the proteins sensitively, but unfortunately fluorescence fades with time and ultraviolet radiation is to be required for detection of the protein bands. In this paper, we describe a mixed dye staining method which takes BBR as a supplement in the established CB staining to detect proteins in polyacrylamide gels rapidly and sensitively. A rapid and sensitive staining method employing a mixed dye technique has been developed. Coomassie Brilliant Blue R-250 (CB, 0.2%) and Bismark Brown R (BBR, 0.05%) were mixed up in the volume ratio of 1:0.75 for staining. In this staining solution, BBR inhibits the binding of CB to gel matrix, and thus increases the staining effect of CB on protein band, and also reduces destaining time. As a result, the mixed dye staining enables the staining/destaining to be completed within 20/25 min and detection of up to 25 ng of bovine serum albumin (BSA). In this method, CB was used for protein staining and BBR for inhibiting the background staining by CB through the counter ion effect. It is noticeable that no changes has been made for the established CB staining method except the use of mixed dye instead of CB.
