Hepadnaviruses의 genome 복제과정은 retrovirus의 복제과정과 유사한 역전사 과정을 거치는데, 두 번의 primer translocation과 한 번의 template switching을 거쳐 partial duplex DNA genome을 형성한다. 이러한 복제과정에서 중추적 역할을 하는 HBV DNA 중합효소는 terminal protein (TP), spacer (SP), reverse transcriptase (RT), RNase H (RH)의 네 개 domain으로 구성되어 있으며, TP domain 의 protein priming으로부터 시작하여 역전사 효소 활성을 가지고 있는 RNA-directed, DNA-directed DNA polymerase인 RT domain이 DNA를 합성하는 동시에 일종의 RNase인 RH domain이 RNA strand를 제거함으로써 RNA template로부터 RNA-DNA hybrid를 거쳐 double strand DNA genome을 합성하게 된다. HBV의 RH domain은 단순히 RNA template를 제거함으로써 복제과정에 참여하며 packaging step에 관여한다고 알려져 있었으나 최근 retrovirus의 RNase H가 strong stop DNA transfer 즉, DNA 합성의 시작단계에 관여하고 있다는 RNase H의 효소 활성과는 별개의 역할이 보고되면서 HBV의 RH의 경우에도 유사한 역할이 기대되었다. 이에 본 연구에서는 RNase H domain에 촛점을 맞추어 clustered charged-to-alanine scanning mutagenesis (CA mutagenesis)를 이용, 효소 단백질의 구조변화를 최소화하는 범위 내에서 단백질상의 주요 아미노산들을 변이 시킴으로써 RNase H domain이 복제에 기여하는 mechanism을 조사하고 각 단계에 관여하는 amino acid residues를 규명하고자 하였다. CA mutagenesis를 근간으로 6개의 mutants를 제작하였고 Southern blot, RNase protection assay, primer extension에서는 흥미롭게도 각 mutant의 phenotype이 다양하였는데 2개의 mutants (CA13, CA15)는 변이에 대하여 tolerance를 가져 wild type과 같은 phenotype을 보였으며 또 다른 2가지의 mutants (CA10, CA14)는 packaging 단계에서 효율이 현저히 떨어지는 결과를 나타내었다. 나머지 두개의 mutants (CA11, CA12)가 DNA 합성 과정에서 defect를 나타내었다. CA11의 경우 replicative intermediates를 조사해 본 결과 minus strand까지만 합성되는 phenotype을 보였다. 이것은 두가지 가능성을 제시하는데 첫째, 두번의 primer translocation 중 plus strand DNA synthesis 시 두번째 translocation 이 제대로 이루어지지 않고 또한 in situ 합성에도 실패하여 그 단계에서 복제의 진행이 멈추었을 경우와 둘째, circularization단계 즉, 세번째 translocation인 template switching이 성공하지 못했을 경우가 그것이다. 다시 말하여 CA11 mutant에서 변이시킨 D777, D778 residues가 이 두가지 step 중 적어도 한가지 단계에 개입되어 있음을 의미한다. CA12의 경우는 single strand DNA (SS), duplex linear DNA (DL), relaxed circular DNA (RC)의 세가지의 replicative intermediates 중에서 오로지 RC DNA만을 합성하지 못하는 결과를 보였다. 즉, CA12 mutant에서 substitution 된 두 arginine residues (R781, R783)가 RC DNA 형성 시 template switch에 작용함을 암시한다. 이상의 연구 결과는 HBV의 RNase H domain 역시 retrovirus의 그것과 마찬가지로, RNA degradation이라는 효소촉매적 기능 외에 polymerase라는 multifunctional protein의 한 domain으로서 다른 domain과의 구조적 상호작용을 통하여 바이러스의 reverse transcription 반응에 적어도 3개의 step에서 작용함을 시사한다.

Hepatitis B virus (HBV), a prototype of hepadnaviruses, is a DNA virus with a circular, partial duplex DNA genome. Hepadnavirus synthesize its partial duplex DNA genome through the reverse transcription via three template switch reactions. HBV DNA polymerase that plays a key role in replication is composed of four subdomains; terminal protein (TP), spacer (SP), reverse transcriptase (RT), RNase H (RH). The first step of reverse transcription is a protein priming reaction by TP domain of HBV polymerase. Then the RT domain of the polymerase which has the reverse transcription activity as an RNA-directed, DNA-directed DNA polymerase synthesizes minus strand DNA, as its associated RH domain degrades the template RNA. In addition its catalytic function, studies have shown that the RNase H domain contributes to packaging step. Moreover, recently, the retroviral RNase H was shown to play a role in strong stop DNA transfer. To further explore the role of HBV RNase H domain, we employed charged-to-alanine scanning (CA) mutagenesis. Using CA mutagenesis, substitution for charged residues can be introduced with minimal local structural alteration. We produced six CA mutants and these mutants were analyzed by Southern blot, RPA (RNase Protection Assay) and primer extension experiments. Two mutants (CA 13, CA 15) showed wild-type like phenotype and another two mutants (CA 10, CA 14) exhibited a defect for packaging step. CA 11 mutant was able to produce minus strand DNA but failed to produce plus-strand DNA. We interpreted this data indicating a defect on the primer translocation process during plus strand DNA synthesis. Also CA 12 mutant synthesized the SS (single strand) DNA and the DL (duplex linear) DNA, but failed to produce RC (relaxed circular) DNA. This result implies that substituted arginine residues (R781, R783) of CA 12 mutant are responsible for the template switch process necessary for RC DNA formation. Collectively, our data suggests that the RNase H domain of the HBV polymerase plays an important structural role as well as its catalytic role during the viral reverse transcription.
