Telomere maintenance is essential for the continued proliferation of dividing cells and is implicated in chromosome stability and cell immortalization. Telomerase activity allows cells to maintain their telomeric DNA and contribute to the indefinite replicative capacity of cancer cells. Telomerase activity is expressed in most type of cancer cells but not in normal somatic cells, suggesting that telomerase may be an important target for cancer chemotherapy. Inhibition of telomerase results in telomere erosion, leading to the subsequent growth arrest of cancer cells followed by senescence or cell death. In this study, extensive propagation of cancer cell line in the presence of DPNS (3-(3,5,-dichlorophenoxy)-nitrostyrene) resulted in progressive telomere erosion followed by the induction of senescence phenotype. Flow cytometric analysis showed that DPNS-treated HT1080 cells were in growth arrest of G2/M phase. To investigate a relation between telomere dysfunction and activation of senescence-signaling pathway which is associated with DNA damage, western blot analysis were performed in DPNS-treated HT1080 cells. Progressive telomere shortening induced by DPNS triggered up-regulation of p53, p21, p27 protein expression and down-regulation of cyclin B protein expression. Therefore, the G2/M arrest observed upon telomerase inhibition may be mediated by p53-dependent pathways.
