(1) 쑥 및 칡의 추출물의 카테킨의 함량을 Iwasa와 Torri의 방법을 변형하여 측정한 결과 네 가지의 추출물 중에서 카데킨의 함량은 쑥의 열수추출물이 8~10mg/100g으로 가장 많았으며 쑥의 에탄올 추출물은 3~4mg/100g, 칡의 경우 열수 및 에탄올추출물의 카데킨 함량이 1~2mg/100ml으로 거의 동량인 것으로 나타났다. (2) 쑥 및 칡의 추출물을 班PLC를 이용해 주요 카레킨의 성분을 비교한 결과 쪽의 열수추출물 및 에탄올추출물과 칡의 열수추출물은 (-)-Epigallocatechin (EGC)의 함량이 가장 높았으나 칡의 에탄올추출물은 (-)-Epicatechin gallate (ECg)의 함량이 가장 높았다. 또한 네 가지의 추출물 모두 (-)-Epicatechin gallate (ECg)가 없었으며 쪽 특유의 카데킨이 칡보다 높은 것으로 나타났다. (3) 시간에 따른 혈중 에탄을 농도를 GC로 측정한 결과 각각 에탄올을 투여한지 1시간 후의 에탄을 농도에서는 CON-E의 경우가 가장 높은 것으로 나타났고 MDW-E, MOH-E, POH-E 그리고 PDW-E 모두 CON-E에 비해 현저하게 감소하는 것을 알 수 있었다. 특히 MDW-E와 MOH-E는 CON-E에 유의하게(p<0.05) 낮은 값을 나타냈다. 또한 에탄올을 투여한지 2시간 후에도 1시간 후와 비슷한 양상을 나타내었는데 MDW-E, MOH-E, POH-E 그리고 PDW-E 모두 CON-E에 비해 현저하게 감소하였으며 MDW-E와 MOH-E는 CON-E에 유의하게(p<0.05) 낮은 값을 나타냈다. (4) 에탄올에 의한 간기능 저하정도를 알아보기 위한 GO, GPT의 시간별 혈중 농도는 각 실험군마다의 유의적인 차이는 보이지 않았으나 GOT 농도의 경우 CON-G에서 5시간이 0시간에 비해 2.5배의 증가를 보인 반면 MDW-G는 1.6배, MOH-G가 0.98배의 증가만을 보였으며 PDW-G와 POH-G는 오히려 0시간에 비해 5시간이 GOT의 농도가 감소하는 경향을 나타냈다. GPT 농도의 경우에는 MDW-G와 MOH-G에서 CON-G에 비해 다소 증가하였으나 PDW-G와 POH-G에서는 감소하였다. (5) 간실질세포를 배양하며 시간에 따른 변화를 현미경으로 관찰한 결과 12시간 배양 후에는 각 군들끼리 세포수나 형태면에서 볼때 커다란 차이는 없었으나 24시간 배양 후부터는 CON-L보다 쑥과 칡추출물을 처리한 군들에서 더 많은 세포수를 보였으며 세포의 모양도 잘 유리됨을 알 수 있었다. 또한 36시간 배양 후 MDW-L에서는 눈에 띄게 세포수가 증가하였다. (6) 간실질세포 배양 36시간 후 배지를 걷어내고 밑면에 모인 세포를 모아 현미경으로 수를 측정하여 평균을 낸 결과는 2% MDW-L가 가장 많았으며 CON-L의 세포수가 가장 적었다. 또한 배양 후 취한 세포내의 GO, GPT의 활성치는 실험군간의 차이가 없이 나타났다.

This study was conducted to investigate the effects of mugwort and Puerarice radix roots extracts on the blood ethanol concentration and liver function in rats. The effects of mugwort and Puerariae radix roots extracts on the blood ethanol concentration was studied in Sprague-Dawley rats (10 weeks old) administrated p.o. with 25% ethanol (5g/1kg body weight) and then injected with mugwort and Puerariae radix roots extracts (at the 2% levels of daily feed consumption compared with the concentration of catechins level in mugwort and Puerariae radix roots extracts) in caudal vein. SD rats were divided into five groups . control group (CON-E, only ethanol and 0.85% saline sol'n treated instead of each extracts), water extracts of mugwort treated to the control (MDW-E), ethanol extracts of mugwort treated to the control (MOH-E), water extracts of Puerariae radix roots treated to the control (PDW-E) and ethanol extracts of Puerariae radix roots treated to the control (POH-E). And then rat plasma of each time (Ohr, 1hr, 2hr, 3hr) was investigated ethanol concentration by gas chromatography. Another rats were measured at the time of 0 and 5hr for the test of GOT(Glutamic Oxaloacetic Transaminase) and GfT(Glutamic Pyruvic Transaminase). Components of each extracts were analyzed by using high performance liquid chromatography. The effects of mugwort and Puerariae radix roots extracts on the liver function were studied in culture of rat hepatocyte composed of nine groups : Control group and eight groups treated with each extracts (1% & 2% MDW, 1% & 2% MOH, 1% & 2% PDW, 1% & 2% POH). Condition of rat hepatocytes cultured for 36hr at 37℃(5% CO_(2) incubator), number of cells, GOT and GPT activity were investigated. The results obtained were summarized as follows ; 1. Catechins level of mugwort and Puerariae radix roots extracts was 8~10mg/100g (MDW), 3~4mg/100g (MOH), 1~2mg/100m1 (PDW, POH) 2. The contents of (-)-Epigallocatechin was high in MDW and (-)-Epicatechin gallate was also high in POH. But (-)-Epicatechin gallate was low in MDW, MOH, PDW and POH. 3. The effects of mugwort and PRE extracts on the blood ethanol concentration were as follows; 1) The order in ethanol degradation efficiency was MDW-E > MOH-E > PDW-E > POH-E > CON-E. 2) Ethanol concentration significantly decreased (p<0.05) in MDW-E and MOH-E. 4. The effects of mugwort and PRE extracts on the liver function were as follows; (rat hepatocytes cultured for 36hr at 37℃) 1) Cells condition of MDW-L was better than other groups. 2) The order in number of cells (rat hepatocytes) was 2% MDW-L > 1% POH-L > 1% MDW-L > 1% PDW-L > 2% PDW-L > 1% MOH-L > 2% POH-L > CON-L. Therefore, this results suggest that the effects of mugwort and Puerariae radix roots extracts may an important role in degradation ethanol and recovery liver function in body.
