In the first study, to increase the release of doxorubicin at a hyperthermic temperature and to prolong the stability of modified liposomes in bovine serum solution, the surface modification of temperature-sensitive liposome was performed with poly(N-isopropyl-acrylamide-co-acrylamide) [poly(NIPAAm-co-AAm)] and polyethylene-glycol (PEG). Four types of liposomes were prepared, and their stabilities and drug release characteristics were studied. To determine the stability of the liposomes, we measured turbidity change, particle size distribution and protein adsorption by UV-spectrophotometer, particle size analysis and fluorescence spectrophotometer, respectively. In addition, drug release from liposomes was measured with respect to temperature and time by fluorescence spectrophotometer. The liposomes modified with poly(NIPAAm-co-AAm) and PEG showed a large amount of drug release and a high level of stability in bovine serum solution. These results suggest that the modification of liposomes using poly(NIPAAm-co-AAm) and PEG, respectively, contribute to reduce protein adsorption and enhance drug release from liposomes in bovine serum solution. We modified the surface of temperature-sensitive liposome with a copolymers of N-isopropylacrylamide and octadecylacrylate, which exhibited a lower critical solution temperature at around 30 ℃. After mixing the surface modified temperature-sensitive liposomes with poly(N-isopropylacrylamide) solution, poly(N-isopropylacryl-amide) solution containing temperature-sensitive liposomes formed a physically cross-linked gel through heating the solution above their lower critical solution temperature. The release of drug from temperature-sensitive liposomes was determined by measuring the fluorescence intensity. The drug release from temperature-sensitive liposomes in poly(N-isopropylacrylamide) gel gradually showed sustained-release with increasing temperature.
