Comparative analysis of human and chimpanzee genome is powerful tool to understand evolutionary features and marker development. As nucleotide sequences of chimpanzee chromosome 22 were recently completed, evolutionary analyses concerning functional genes and mobile genetic elements between human chromosome 21 and chimpanzee chromosome 22 were available. (1) KAP13 genes located on human chromosome 21q22 were analyzed using various molecular tools including their orthologous loci of ape and monkey genomes. PCR amplification was performed using genomic DNAs of primates (1 human, 1 chimpanzee, 1 gorilla, 1 orangutan, 8 gibbons, 3 Old World monkeys, and 2 New World monkeys) and two different primer sets, indicating that KAP13.4 gene was not detected in the chimpanzee and gorilla, whereas the bands were appeared in human, orangutan, gibbons, and monkeys. The PCR products of KAP13.4 gene were sequenced and analyzed by GCG, GENETYX and MEGA2 programs. Noticeably, sequences of AGC AGC CTG GTC (amino acids SSLV) were deleted in monkey species only. The HU-KAP13.4 sequences indicated 33 site differences and had 86-96% identity with gibbons and monkeys on their open reading frames. Within monkeys, they showed identical sequences each other, whereas agile gibbon and white-handed gibbon showed one transition mutation. (2) Human specific Alu elements were also investigated by comparative analysis between human chromosome 21 and chimpanzee chromosome 22. On human chromosome 21q22 (accession no. AL163282), new Alu Yj4 element was identified by bioinformatic tools. Using various human DNAs, PCR analysis was performed on Alu Yj4 locus, indicating that no polymorphism was detected. It seems to be proliferated approximately 3.66 - 6.87 Myr ago in human genome only. Furthermore, Alu Yj4 family was investigated in human whole genome using Alu Yj4 sequences (accession no.
