MALDI mass spectrometry 분석결과, halocidin은 3453.25Da의 분자량을 지닌것으로 확인되었고, 18개의 아미노산을 지닌 펩타이드 (18mer)와 15개의 아미노산을 지닌 펩타이드 (15mer)가 하나의 이황화 결합으로 연결된 이량체 분자 구조를 가지고 있었다. 이황화결합은 18mer의 12번 cysteine 잔기와 15mer의 9번 cysteine 잔기 간에 이루어져 있으며, 18mer의 4-18번의 아미노산 서열은 15mer의 1-15번의 아미노산 서열과 완전히 일치했다. 본 연구에서는 halocidin을 구성하는 15mer와 18mer의 펩타이드를 각각 합성하여 이들을 다시 산화시켜 이황화 결합을 만들어 냄으로써 15mer의 dimer (15dimer), 18mer의 dimer (18dimer) 및 natural halocidin과 같은 구조의 15-18mer의 dimer (heterodimer)인 세 종류의 dimer를 제작하였다. 이들이 지닌 항균활성을 그람양성균과 그람음성균에 대해 분석한 결과, monomer들의 항균활성보다 dimer들의 활성이 더 강하였고, 세 종류 dimer들의 항균활성에 있어서는 18dimer, heterodimer, 15dimer의 순으로 크게 나타났다. 또한 본 연구에서는 천연 펩타이드인 halocidin보다 구조가 작으며 안정하고 항균활성이 유지되는 이상적인 구조의 펩타이드를 찾아내기 위하여 다양한 펩타이드들을 design 하였는데 즉, 18mer의 C-말단으로부터 하나씩의 아미노산이 잘려진 펩타이드들 (17mer, 16mer)과 18mer의 7, 8번 histidine이 lysine으로 치환된 펩타이드 (18mer K)가 먼저 제작되어 연구되었다. 이제까지의 실험결과에 의하면 18mer K의 dimer가 가장 항균활성이 컸으며, 절단한 펩타이드들의 dimer들은 heterodimer와 거의 비슷한 항균활성을 지니고 있었다. 사람 적혈구에 대해 펩타이드들이 지니는 용혈활성 (hemolysis activity)을 측정한 실험에서는 halocidin, 18dimerK와 monomer들은 거의 용혈작용을 일으키지 않았고, dimer들 (18dimer, 17dimer, 16dimer)은 약간의 용혈활성을 나타냈다. 그러나 이들의 용혈활성도는 대조군으로 사용한 기존 항균 펩타이드인 melittin과 clavanin AK보다는 훨씬 낮았다. 따라서 이제까지의 연구결과, halicidin은 항균활성이 우수하고 세포 독성이 최소화된 이상적인 펩타이드 항생제를 개발하기 위한 적합한 구조를 가지고 있는 것으로 판단되며, 따라서 앞으로의 연구는 halocidin의 구조와 항균활성간의 관계를 밝힘으로써 새로운 천연 펩타이드 항생제를 개발하는 방향으로 진행될 것이다.

Halocidin consisted of 18 amino acid peptide (18mer) and 15 amino acid peptide (15mer) connected by disulfide bond, which formed between 12th cysteine residue of 18mer and 9th cysteine residue of 15mer. 4-18 residues of 18mer amino acid sequence were completely identical to 1-15 residues of 15mer amino acid sequence. In this study, we prepared a variety of congeners (15mer, 18mer, 17mer, 16mer, 18mer K) of synthetic halocidin deduced from the natural form. Several dimers (15dimer, heterodimer, 18dimer, 17dimer, 16dimer, 18dimer K) were also produced from dimerization of synthetic halocidin performed according to air oxidation methods. In case of 18 dimer K, it was replaced two histidin residues with lysines. Other two congeners, 16mer and 17mer, were C-terinally truncated in 18mer of halocidin by two and one residue, respectively. We tested the antimicrobial activities of synthetic peptides against Gram-positive and Gram-negative bacteria. Dimer forms of halocidin congeners were more active than monomers against Gram-positive and Gram-negative bacteria. The most active form was 18dimer K followed by 18dimer, heterodimer. In hemolysis test, monomers were not hemolytic activity. dimers were only modestly so.
