The study was conducted to investigate the effect of onion extract on pear browning. Pear browning is caused by the enzymatic oxidation of phenolic compounds by polyphenol oxidase(PPO, o-diphenol: O_(2) oxidoreductase, EC 1.10.3.1). PPO of pear was purified by ammonium sulfate precipitation (30-70%) and dialysis. The enzyme was purified to 1.42 fold and the yield was 31.1 %. The optimum conditions for the enzyme reaction were pH 6.0-7.0 and temperature of 30-40℃. The enzyme was most stability at pH 6.0 and 70℃. The pear PPO was most active with 4-methylcatechol as a substrate, followed by catechol, (+)-catechin, whereas the enzyme showed poor substrate, specifies toward resorcinol and hydroquinone. Dithiothreitol markedly inhibited PPO activity and L-ascorbic acid, L-cysteine, 2-morcaptoethanol and sodium pyrosulfite also inhibited PPO activity. Heating onion extract exhibited higher inhibitory effect on the pear PPO activity than that of the fresh one. The pear slice showed a rapid change to a brown color, whereas that immersed in fresh or heated onion extract reduced the intensity of the brown color formation. Regardless of substrate used, the heated onion extract exhibited more inhibitory effect on pear PPO than that of the fresh onion extract. The inhibitory effect of onion extract was dependent upon heating temperature and reaction time.
