오늘날 심각한 문제로 대두되고 있는 약물 내성률의 증가와 기회성 진균 감염의 증가를 막기 위해서는 올바른 항균제 사용이 이루어져야함과 동시에 기존 항균제의 독성, 내성 문제 등의 단점을 보완할 수 있는 천연 항균제의 개발이 필요하며 이에 대한 다양한 연구가 친행되고 있다. 본 연구는 항균 작용을 가지는 정유의 자원으로 잘 알려져 있는 Thymus속 식물 중 국산 자원 식물인 백리향 및 섬백리향의 정유를 추출, 분석하여 병원성 진균(Aspergillus,Candida species, Trichophyton rubrum, Trichophyton mucoides, Cryptococcus nwoformans, Blasto-schizomices capitatus)과 항생제 내성을 갖는 병원성 세균(Streptoco-ccus pneumoniae, Staphylococcus aureus, Salmonella enterjtidis, Salmonella typhimurium)에 대한 항균력을 액체 배지 희석법과 디스크 확산법을 통해 확인하고 정유와 기존 항균제와의 병용 효과를 checker-board microtiter assay를 통해 관찰하였다. 또한 생쥐에 질 칸디다증을 유발시켜 백리향 및 섬백리향 정유의 in vivo 항진균 효과를 실험하였다. 연구 결과 백리향 및 섬백리향 정유의 GC-MS 분석에 의해 주성분인 thymol 함량이 각각 41.7%, 39.8%로 나타났다. 백리향 및 섬백리향 정유의 8종 진균에 대한 억제 효과 실험 결과 MICs와 MFCs가 0.04-0,78㎎/㎖로 유의성 있는 억제를 보여주었으며 T.rubrum 에 대한 억제 효과가 가장 강했다. T. rubrum 에 대한 백리향 및 섬백리향 정유와 ketoconazole의 병용 실험에서 FICI가 0.35-0.37로 유의성 있는 상승 작용을 나타냈다. 백리향 및 섬백리향 정유의 4종 세균에 대한 억제 효과 실험 결과 S.pneumoniae가 가장 높은 감수성을 보여 MICs가 0.125-10㎎/㎖ 였으며, 특히 내성균 4059에 대한 백리향 정유의 M가 0.125㎎/㎖로 가장 억제 작용이 높았다. 백리향 및 섬백리향 정유와 항생제의 병용으로 S. pneumoniae 에서는 감수성균보다 내성균에 대한 상승 효과가 컸으며, 특히 S. aureus 에서 FICI가 0,09 - <0.38로 가장 높은 상승 작용을 나타냈다. 섬백리향 정유의 C.albicans 억제 작용에 대한 in vivo 실험 결과 섬백리향 정유는 약물 투약에 따른 항진균 효과가 clotrimazole보다 약했지만, 투약 종료 시점에서는 모두 균 성장이 관찰되지 않았다. 백리향과 섬백리향 정유의 in vitro, in vivo antimicrobial activity실험 결과 이들 정유의 강한 antimicrobial activity를 확인하였고 또한 기존 항균제와의 병용에 의한 높은 상승 효과를 확인할 수 있었다. 이러한 결과는 한국산 Thymus 속 자원 식물인 백리향, 섬백리향 정유 또는 주성분인 thymol과 기존 항균제와의 병용에 의한 상승 호과에 의해 항균제 사용을 줄일 수 있는 의약품으로의 개발 가능성을 시사하며, 이들 정유의 내성균에 대한 억제 효과 및 작용 기전에 대한 심층적인 연구로 내성균 치료제 개발의 기초 자료가 될 수 있으리라 사료된다.

The necessity of new natural antimicrobial agents for overcome toxicity and resistance of existing antimicrobial agents is growing up and proper use of antimicrobial agents need to prevent of drug resistance and opportunistic fungal infection, which is rising serious problem at present. Also the many study about development of natural antimicrobial agents has been carried out. In this study essential oils from Thymus quinquecostatus and T.magnus, which are species native to Korea and Thymus species are well known as antimicrobial natural sources, were extracted and analyzed. The antimicrobial activities of essential oils from two Thymus species were evaluated against common pathogenic fungi(Aspergillus, Candida species, Trichophyton rubrum, Trichospo -ron mucoides, Crytococcus neoformans, Blastoschizomyces capitat -us) and susceptible or resistance pathogenic bacteria(Streptococc -us pneumoniae, Staphylococcus aureus, Salmonella enteritidis, Sal -monella typhimurium) by broth dilution method and disk diffusion test. The-combination effects of two Thymus oils with antimicrobial agents were tested by checkerboard microtiter assay. Furthermore, in vivo antifungal activity of two Thymus oils were tested by experimental vaginal candidiasis. As the results of analysis by GC-MS, thymol which is main component of T. quinquecostatus and T, magnus oils were 41.7% and 39.8%. In antifungal activity tests against 8 species of fungi, two Thymus oils showed significant inhibition with MICs and MFCs in the range of 0.04-0.78 mg/ml and T, rubrum was most susceptible. Additionally, the two Thymus oils exhibited significant synergism with ketoconazole against T. rubrum, resulting in the same fractional inhibitory concentration indices (FICI) of between 0.35 and 0.37. In antibacterial tests against 4 species of bacteria, S. pneumoniae was most susceptible and the two Thymus oils showed signigicant inhibition with MICs in the range of 0.125-1.0mg/ml. Especially, T, qurnquecostatus oil was most effective with MIC in 0.125 mg/ml against resistant bacteria 4059. Additionally, the two Thymus oils exhibited significant synergism with antibiotics against resistant S. pneumoniae. Especially the highest synergism was shown against S. aureus resulting in FICI of between 0.09 and 0.38. In vivo tests, antifungal activity of T, magnus oil accompany by medication was milder than clotrimazole but growth of fungi at the end of medication had not seen both. High antimicrobial activities of two Thymus oils were identified jn vitro & in vivo antimicrobial activity tests. Additionally, synergistic effects of two Thymus oils were identified in combination with antibiotics. These result exhibit potentialities of development of drug in reduce amount of antibiotics by synergistic effect in combination with antibiotics and two Thymus oils or thymol. The deep study about growth inhibition against resistant bacteria and its mechanism of two Thymus oils will be the fundamental data for development of therapeutic agents against resistant bateria.
