Numerous DNA detection methods using fluorescence, visible organic dye, silver and radio-labeling have been proposed in electrophoresis gels. The widely used method for staining of DNA is to use the fluorescence-intercalating agent such as ethidium bromide with UV irradiation. However, it has disadvantages due to the strong mutagenic effects of EB. In this study, instead of EB staining, quick, sensitive and safe methods for detection of DNA in agarose and polyacrylamide gels were developed. The counterion-dye staining technique using indoine blue and methyl orange was applied for the staining of DNA in agarose gel. Furthermore, in IB/MO dye staining, the ‘peel off’ effect that dried gel peels off from filter paper after gel drying increased the contrast between DNA bands and gel background. IB/MO dye staining method can detect as little as 5 ng per band of λ DNA/HindⅢ. This staining method enhanced sensitivity and decreased gel background. In polyacrylamide gels, highly sensitive detection of nucleic acid in picogram range has been achieved by silver staining. It increased sensitivity by using eriochrome black T as a silver ion sensitizer. The detection limit of EBT-silver staining method was 1-4 pg per band of ΦX174 DNA/HaeⅢ. Especially, in low base pair DNA, the sensitivity was more sensitive than previously published silver staining methods.
