Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) is one of the most widely used techniques for the separation and identification of the proteins from complex mixtures. Base on this tool, protein samples separated by 1- or 2-D SDS-PAGE are visualized by various staining methods using visible organic dyes, fluorescent dyes, and so on. Despite many advantages, other disadvantages limit its use for high-throughput proteomics. Here, we introduced several easy, rapid and sensitive protocols to detect total proteins and phosphoproteins in SDS-polyacrylamide gel. First, an advanced negative staining method using Phloxine B (PB) comparable to silver staining was described. It allowed the detection of proteins down to 0.1~0.8 ng within 50 min. Comparing with Imidazole-zinc (IZ) staining, the most common negative staining method, PB staining provided higher sensitivity and better contrast between the protein bands and gel matrix. In addition, we also studied to identify phosphoproteins in gel, which is useful in the fields of biochemistry and molecular biology. Fluorescent staining methods using Anthracene Chrome Red A (ACRA) or 5-Chloro-8-hydroxyquinoline (CHQ)-SYPRO Ruby was developed respectively. They were able to form ternary complexes (dye-Al3+-phosphoprotein) in the gel matrix contributed by the metal chelating property of dyes and the affinity of aluminum ion to the phosphate groups on the proteins. The method using ACRA was negative staining based on fluorescence differences for phosphoprotein and non-phosphorylated protein. ACRA staining was able to detect 1~2 ng of α-casein and β-casein, 8~16 ng of ovalbumin (OVA) and κ-casein within 130 min. The ACRA staining method had high sensitivity compared to Pro-Q diamond staining and showed better selectivity than that. In CHQ-SYPRO Ruby staining, since phosphoproteins –CHQ complex showed green fluorescence distinctly different from red fluorescence of non-phosphoproteins and SYPRO Ruby complex, this detection method allowed to monitor phosphoproteins with high selectivity. These novel staining methods were simple and sensitive, in addition, CHQ-SYPRO Ruby staining also allowed high specificity for phosphoproteins.
