In an attempt to investigate the properties of the malignant progression of glioma cells, we examined the correlation between cell migration and glial cell-derived neurotrophic factor (GDNF) secretion of two glioma cell lines which differ in their invasive phenotypes. Here, we show that the high-grade C6 cells are more migrative and secrete more GDNF than the low-grade Hs683 cells, suggesting that the migrative capacity of glioma cells may be linked to the GDNF production. Treatment of the Hs683 cells with GDNF significantly increased migration comparable to the C6 cells as evidence by transwell migration assay and wound migration assay. The result reveals an autocrine and/or paracrine effect of GDNF on promotion of the glioma cell migration. To elucidate a molecular mechanism for the effect of GDNF, we examined the involvement of mitogen-activated protein kinases (MAPKs) including c-Jun N-terminal protein kinase (JNK), extracellular signal-regulated kinases (ERKs) and p38 MAPK in Hs683 cell migration induced by GDNF. A prominent activation of JNK, ERKs and p38 MAPK was observed in Hs683 cells treated with GDNF. By using chemical inhibitors and dominant negative (DN) mutant constructs of these MAPKs, we showed that the activation of JNK, ERK-1/2 and p38 MAPK was critical for Hs683 cell migration induced by GDNF. Our findings revealing a molecular mechanism for the promoting effect of GDNF on glioma cell migration may contribute to the development of therapeutic interventions to prevent the malignant progression of human gliomas. Human tumors frequently exhibit constitutively activated Ras signaling which contributes to the malignant phenotype. We previously showed that p38 MAPK is a key signaling molecule differentially regulated by H-ras and N-ras, leading to H-ras-specific cell invasive and migrative phenotypes in MCF10A human breast epithelial cells. In this study, we further investigated the role of p38 MAPK pathway in the induction of metastatic potential in MCF10A cells as a "gain of function" study. First, we tested whether the activation of p38 MAPK can confer invasive and migrative abilities in MCF10A cells which were originally non-invasive and nonmigrative. A significant role for p38 MAPK in cell invasion is further supported by the observation that p38 MAPK activation by constitutively active mutant MAP kinase kinase (MKK)-6 transfection is sufficient to induce invasive and migrative phenotypes in MCF10A cells. Activation of p38 MAPK MAPK pathway by transfecting MKK6 caused a selective up-regulation of MMP-2. We also studied to elucidate the transcriptional regulation of MMP-2 by p38 MAPK pathway leading to the invasive and migrative phenotypes of MCF10A breast epithelial cells. By using 5' deletion mutant constructs of MMP-2 promoter, we showed that deletion of the AP-1 site spanning -1591 bp to -1259 bp caused the greatest reduction of MMP-2 promoter activity both in MKK6-and H-Ras-activated MCF10A cells, suggesting that the AP-1 binding site is critical for the MMP-2 promoter activation. DNA binding and transcriptional activities of AP-1 were increased by MKK6 or H-Ras as evidenced by EMSA and luciferase assay using a AP-1-driven plasmid. By performing an immunoinhibition assay, we revealed the activating transcription factor (ATF)2, which was activated in MKK6- and H-Ras MCF10A cells, as a transcription factor for MMP-2 gene expression through binding to the functional AP-1 site. Activation of ATF2 by MKK6 or H-Ras was crucial for MMP-2 promoter activity as well as induction of invasive and migrative phenotypes in MCF10A cells. This is the first report revealing ATF2 as an essential transcription factor linking MKK3/6-p38 MAPK signaling pathway to MMP-2 up-regulation, providing evidence for a direct role of ATF2 activation in malignant phenotypic changes of human breast epithelial cells.
