At an initial stage of this study, a conjugate between colloidal gold and protein A, a binding protein to antibody (particularly, immunoglobulin G), was used as a signal generator. However, since protein A bound all antibodies regardless of their specificities, the sensitivity of the system was lower, due to competitive bindings among them, than that of the conventional system, ELISA. In order to increase the sensitivity, we then investigated a signal generator that recombinant HIV antigens (p24, gp41, and gp36) were conjugated to a colloidal gold with a considerate adjustment of their solubilities in a less amount of urea. If this conjugate was used in the assay, the antibodies, specific to the antigens, present in a sample cross-linked the conjugate and those immobilized on the signal generation pad. This approach significantly increased the sensitivity, resulting from a selective binding of the conjugate to the antibodies raised against the HIV antigens and also detection of the antibodies of immunoglobulin (Ig) M class in addition to IgG. Furthermore, two analysis test lines were formed in the test device such that two different HIV subtypes, type 1 and type 2, were separately detected at the same time. The new analytical system can be used for rapid analyses of HIV infections with an enhanced sensitivity in emergency rooms and private hospitals as a substitute of ELISA.
