The phosphate solubilizing bacteria, HB-15 and HB-22 possessing a strong ability to solubilize insoluble phosphate, were isolated from cropland soil. These bacteria oxidized glucose to gluconic acid and sequentially to 2-ketogluconic acid, which highly solubilized hydroxyapatite into orthophosphate. Based on DNA sequence of a conserved partial 16S rRNA gene, HB-15 and HB-22 were identified as Psedomonas aeruginosa and Pantoea spp, respectively. At the same time, these bacteria showed significant antifungal activity against several plant pathogens, such as Phtophthora capsici, Rhizoctonia solani and Fusarium oxysporum in an agar medium. To isolate antifungal compounds from culture filtrates of these bacteria, various chromatographic procedures were performed. HPLC and GC-MS results confirmed the compounds produced by HB-15 and HB-22 as phenylacetic acid and 2-aminobenzoic acid, respectively. These compounds showed antimicrobial activities against various bacteria and fungi in vitro test. To investigate protective effects of antagonistic HB-15 in relation to biological control of Phytophthora blight in pepper caused by P. capcici, pathogeniesis-relatetd (PR) proteins and plant defense enzymes related to cell wall lignification were examined in pepper plant preinoculated with HB-15 against a pathogenic strain, P. capsici. Root mortality was 76% in P. capsici treated plants, while it was significantly reduced in HB-15 treated plants. The activities of PR proteins (chitinase and β-1,3,-glucanase) and lignification-related enzymes (peroxidase and phenylalanine ammonia lyase) decreased in root of HB-15 + P. capsici treated plants, but increased in leaves with time. This fluctuation was much greater in P. capsici treated plants compared to HB-15 + P. capsici treated plants although P. capsici treated plants had severer damage. In addition, HB-15 treatments enhanced plant growth and P uptake when compared to control. In conclusion, HB-15 not only promoted pepper growth and P uptake, but also protected pepper against phytophthora blight. To investigate whether Pantoea spp. HB-22 can induce resistance, we tested biocontrol effect against angular leaf spot caused by Pseudomonas syringae pv. lachrymans in cucumber. Additionally, changes in activities of defense enzymes involved in reinforcement of cell walls and PR proteins in leaves of cucumber were investigated. The spatial separation between HB-22 and pathogen in the phyloplane excludes the possibility of direct antagonism. Disease severity was higher in P. syringae pv. lachrymans treated plants, compared to HB-22 treatment. The activity of pathogenesis-related (PR) proteins, such as β-1,3-glucanase and chitinase was increased in P. syringae pv. lachrymans treated plant leaves. However, the increase of PR proteins by pathogen infection was alleviated in HB-22 treated plant. The POD and PAL activity also showed similar increase pattern compared to PR proteins. These results suggested that the increase in PR proteins, POD and PAL activities was a symptomatic response, but not related to plant resistance against the pathogen infection in this study. The effect of biocontrol agents, Pseudomonas aeruginosa HB-15 and Pantoea spp. HB-22 applied individually and in combination was inverstigated against damping-off of caused by Rhizoctonia solani in cucumber plants. Fresh weights and P uptake in bicontrol agents treated plants were significantly higher both individual (HB-15 and HB-22) and mixture (HB-15 + HB-22) treatment than control in shoots. Root mortality was rapidly increased in control compared to biocontrol agents treated plants. NH3-NH4+ and proline concentrations were higher in the control, while they are alleviated in biocontrol agents plants. Induced disease resistance pathway by biocontrol agents was identified comparing to gene expressions induced by salicylic acid (SA) and jasmonic acid (JA). POX and PAL1 expression were increased in both SA and Pantoea spp. HB-22 treated plants. However, P. aeruginosa HB-15 did not increase any gene expression. These results show that combination or individual of P. aeruginosa HB-15 and Pantoea spp. HB-22 can be used as biocontrol agent and biofertilizer.
