우리나라 주요 염생식물에서 생리활성이 우수한 자원을 찾고자 33종 39점을 대상으로 페놀성 화합물의 함량, DPPH free radical에 대한 전자공여능, superoxide dismutase(SOD) 유사활성, 아질산염 소거능을 조사하였다.
염생식물들의 페놀성화합물의 함량은 해당화 줄기 부위가 593.2 μg/㎎으로 가장 높았으며 달맞이꽃 전초 221.7 μg/㎎, 해당화 뿌리 185.2 μg/㎎, 구절초 전초 167.9 μg/㎎, 좀보리사초 전초 141.5 μg/㎎, 갯까치수영 전초 119.7 μg/㎎, 해당화 잎 117.1 μg/㎎, 사철쑥 99.4 μg/㎎로 페놀성 화합물의 함량이 많았다.
DPPH free radical에 대한 전자공여능은 해당화 뿌리의 낮은 농도(SC_(50):10.1㎍)에서 항산화 활성을 나타내었다. 해당화 부위별로 SC_(50) 값은 뿌리, 줄기, 잎, 열매순 이었다.
Superoxide dismutase(SOD) 유사활성은 갯까치수영 전초 추출물이 22.36%로 가장 높은 SOD유사활성을 나타냈으며 다음은 실망초 전초 추출물이 20.09%, 해당화 줄기 추출물이 19.36%, 해당화 잎이 19.24%이었다.

주요 무기질은 인(P)의 함량이 278.8 ㎎%로 가장 높았으며, 칼슘(Ca) 251.1 ㎎%, 철(Fe)이 149.5 ㎎%, 칼륨(K) 63.1 ㎎%, 마그네슘(㎎) 54.6g ㎎%로 비교적으로 높은 함량을 보였으며, 나트륨(Na) 6.3 ㎎%, 구리(Cu) 0.4 ㎎%, 아연(Zn) 0.8 ㎎%가 각각 함유되어 있었다.
또한, 주요 지방산 조성은 stearic acid가 37.56%로 가장 높았으며 다음으로 palmitic acid 20.34%, linoleic acid 11.01%, oleic acid 7.61%등으로 검출 되었으며 이중 불포화 지방산이 20.02% 정도였다.
해당화 뿌리에서는 유리아미노산의 함량이 422.46 ㎎%이었으며, 이중 필수아미노산은 15.13 ㎎%로 전체 유리아미노산의 3.6%이었으며 arginine이 210.99 ㎎%, asparagine 97.92 ㎎%으로 전체 유리아미노산의 약 23.1%를 차지하였다.
유리당 조성 및 함량은 sucrose가 3.33%, fructose 1.29%, glucose 1.26%이었다.
해당화 뿌리의 추출용매에 따른 항산화 활성을 비교한 결과 해당화 뿌리를 11종의 용매를 달리하여 추출한 추출물의 수율은 열수 환류추출시 13.84%로 가장 높았으며 methanol 추출 6.54%, ethanol 추출 4.06%의 추출율을 보였다. 또한, 해당화 뿌리는 추출용매별로 free radical 소거능이 달라 ethanol 추출물, methanol 추출물, 열수 추출물, acetone 추출물등은 대조구로 이용한 α-tocopherol 및 BHT등과 비슷한 높은 free radical 소거능을 보였다.
3종 해당화 뿌리 추출물로 Bacillus cereus, Staphylococcus aureus, Listeria monocytogenes, Escherichia coli 그리고 Salmonella typhimurium등을 대상으로 항균활성을 비교 하였다. 열수 추출물, ethanol 추출물, methanol 추출물을 이용한 항균 활성은 추출물의 처리농도가 증가함에 따라 항균 활성이 증가하였으며 3종 추출물 모두 benzoic acid와 유사하거나 높은 항균 활성을 가졌고 열수 추출물 > ethanol 추출물 > methanol 추출물순으로 항균 활성이 높았다.

해당화 뿌리 ethanol 추출물을 식용 어유에 첨가하여 항온 저장하면서 기존 항산화제와 비교 검토한 결과 해당화 뿌리 ethanol 추출물 첨가구 및 α-tocopherol, BHT 첨가구 모두 산가, 과산화물가, TBA가의 증가가 무처리구 보다 다소 낮은 경향을 보여 어유의 저장 중 산패방지에 효과가 있었으며 유도기간을 연장시켰다.
이상의 결과를 바탕으로 항산화 활성이 뛰어난 해당화 뿌리는 우수한 항산화 소재로의 이용 가능성을 보여주었다. 따라서, 식품에 적용하기 위해서는 추후 각종 독성 실험등 안전성에 관한 연구가 필요할 것이라 사료된다.

This study was carried out to evaluate the phenolic compounds, DPPH radical scavenging activity and nitrite-scavenging ability in methanol extracts from 33 salt marsh plants, and their physiological activities were compared. Among methanol extracts of 33 kinds of salt marsh plants the highest content of polyphenol was in the Rosa rugosa T. stem (593.2 ㎍/㎎). The highest DPPH radical scavenging activity was obtained from Rosa rugosa T. roots (SC_(50):10.1 ㎍). SOD-like activity in the extract of Lysimachia mauritiana whole plant was 22.36%, which was the highest value among tested samples and the extracts of Erigeron bonariensis whole plant, Rosa rugosa T. stem and Rosa rugosa T. leaf were also showed relatively high activities. The leaf of Rosa rugosa T. showed the highest nitrite-scavenging activity among methanol extracts of salt marsh plants.
Rosa rugosa T. root was selected for further study because of the high antioxidative activity among salt marsh plants. The root was analyzed for chemical compositions such as proximate composition, mineral contents, fatty acid compositions, free amino acid compositions and free sugar contents. The results showed that moisture, crude lipid, crude protein, ash, and carbohydrate contents of Rosa rugosa T. roots were 8.83%, 0.41%, 5.89%, 3.81% and 81.06%, respectively. In case of mineral components, Rosa rugosa T. root was rich in P (278.8 ㎎%), Ca (251.1 ㎎%) and Fe (149.5 ㎎%). Various fatty acid components in Rosa rugosa T. roots were identified and the major fatty acids were stearic acid (37.56%), palmitic acid (20.34%), linoleic acid (11.01%), and oleic acid (7.61%). Content of total free amino acids in Rosa rugosa T. roots was 422.46 ㎎%, and the major amino acids were arginine (210.99 ㎎%), and asparagine (97.92 ㎎%). Major free sugars of Rosa rugosa T. roots were sucrose (3.33%), fructose (1.29%) and glucose (1.26%).
Antibacterial activities of hot water, EtOH and MeOH extracts from Rosa rugosa T. roots were investigated against various foodbone pathogens. Hot water extracts had the highest antimicrobial activities against various foodbone pathogens such as Staphylococcus aureus, Esherichia coli, Salmonella typhimurium, Bacillus cereus and Listeria monocytogenes.
Among the various extracts of Rosa rugosa T. roots using 11 solvents, ethanol extract showed relatively high antioxidative activity and desirable extraction yield. The extract was dissolved in water and sequentially fractionated with n-hexane, chloroform, ethyl acetate, butanol and the highest radical scavenging activity was found in ethyl acetate fraction. The fraction was further fractionated through silica gel-adsorption column chromatography, and high performance liquid chromatography (HPLC) to purify antioxidation-active substances. Chemical structures of the purified substances (compounds 1 and 2) were determined by electron-impact ionization mass spectrometry (EI-MS) and nuclear magnetic resornance (NMR) spectroscopy. These substances were identified as catechin and procyanidin. The SC_(50) values of the compound 1 and 2 were 1.56 ㎍ and 4.29 ㎍, and their contents in Rosa rugosa T. roots were 24.02 ㎎/100 g and 7.46 ㎎/100 g dry weight, respectively.
