GLP-1 작용제의 안정성은 GLP-1 리간드를 DPP-IV 가 함유된 것으로 알려진 소태아혈청을 첨가한 배지에 일정시간 처리한 후 루시퍼레이즈 활성도를 측정하여 분해되지 않고 남아있는 작용제를 유추하는 간접방법을 사용하였다. 결과: 각각의 GLP-1 작용제의 용량 반응 곡선을 그려본 결과 xGLP-E4 융합 펩타이드가 GLP-1보다 활성도가 높은 결과를 보여주었다. Background and Purpose: Glucagon like peptide-1 (GLP-1) is an incretin hormone derived from the proglucagon gene at the intestinal L-cells. In the pancreas, GLP-1 stimulates meal-induced insulin secretion, promotes insulin biosynthesis, and improves insulin sensitivity. Because of its insulinotropic activity, GLP-1 has been considered as a good candidate drug for the treatment of diabetes mellitus. However, the clinical use of GLP-1 has been limited by its short half life due to rapid degradation by dipeptidyl peptidase-IV (DPP-IV). The purpose of this study is to evaluate the potency and stability of newly designed GLP-1 analogues based on the amino acid sequence of Xenopus GLP-1, exendin-4, and human glucose-dependent insulinotropic polypeptide (GIP). Methods: The receptor cDNA was co-transfected into HEK293 with cAMP-dependent protein kinase A (PKA)-specific reporter (CRE-luciferase; abbreviated as CRE-luc). Various concentrations of chimeric GLP-1 agonists were treated to the transfected cells for 6 hours and luciferase activities were determined. The stability of agonists was evaluated by determining the activity of the agonists that were preincubated in the presence of fetal bovine serum (FBS) which naturally contains DPP-IV activities. Results: GLP-1 was able to induce CRE-luc activities. A chimeric GLP-1 analog with the sequence of Xenopus GLP-1 and exendin-4 (termed xGLP-E4) was as effective as exendin-4 (EC50; 0.17 nM, 0.11 nM, and 0.44 nM for xGLP-E4, exendin-4, GLP-1, respectively). When GLP-1 was incubated with FBS for 72 hours, the potency was rapidly decreased by 15% of initial activity, but the activity of xGLP-E4 was maintained up to 85% of initial activity. The chimeric peptides with GLP-1 and GIP activated both the GLP-1 receptor and GIP receptors. Conclusions: The new peptide produced by addition of C-terminal ten amino acid sequences from exendin-4 to Xenopus GLP-1 may improve the potency to activate the GLP-1 receptor. Substitution of serine for alanine at the second position of the GLP-1 improved the stability of the peptide against the action of DPP-IV. Combination of GIP and GLP-1 sequences activated both GLP-1 and GIP receptors.
