Growth and glucosinolate content of the root were greatly affected by cultural conditions. The effect of basal medium on growth of wasabi roots was in sequence of mNB, WPM, Anderson, White, 1/4MS, and B5 and that on sinigrin production was in the order of mMS, White, WPM, Anderson, mNB, 1/4MS, and B5. The effect of initial sucrose concentration (i.e. 10, 30, 50, 0 and 100g/L) was investigated in root suspension culture of wasabi. The final root fresh weight increased from 0.445g to 1.448g/L with an increase of initial sucrose concentration from 10 to 70g/L, while 100g/L sucrose repressed the root growth. The highest sinigrin production was achieved at an initial sucrose concentration of 70g/L. Wasabi roots cultured on mNB medium supplemented with various concentration of auxins (IBA, IAA, or NAA: 1, 3, 5, 7, or 10mg/L).
