HCV NS5B protein에 활성이 조절되는 aptazyme을 개발하기 위해 random self-cleaving aptazyme pool을 SELEX방법을 이용하여 NS5B aptamer와 NS5B protein의 binding energy를hammerhead ribozyme의 활성화 구조로 변화 시켜줄 수 있는 communication module을 선별하였다. 이렇게 선별된 self-cleaving aptazymes을 이용하여 NS5B 농도별 활성과 시간별 활성을 조사함으로써 aptazyme의 활성이 NS5B 의존적임을 검증하였다. 검증된 self-cleaving aptazyme을 이용하여 HCV IRES targeting trans-cleaving aptazyme으로의 활성을 검증해 본 결과, 선별된 aptazyme들이 trans-cleavage 활성은 없음을 확인하였다. 이렇게 선별된 self-cleaving aptazymes은 HCV diagnostic sensor로서 활용할 수 있을 것이며, trans-cleavage 활성이 없기 때문에 직접적인 IRES 표적은 할 수 없지만, cleavage되어져 나오는 fragment에 HCV replication을 저해할 수 있는 sequence를 삽입함으로써 항HCV 제제로 사용할 수 있을 것으로 생각된다.

Since the RNA dependent RNA polymerase (NS5B) of HCV plays a central role in the viral replication and highly conserved Internal ribosomal entry site (IRES) of HCV plays a central role in viral protein translation, both are good targets for anti-HCV agents. I developed HCV IRES-targeting hammerhead ribozyme which activity is alloterically regulated by HCV NS5B protein. To develop NS5B-dependent allosteric hammerhead ribozyme (aptazyme), I performed in-vitro selection to identify communication module that can deliver configuration change of NS5B aptamer via binding of NS5B to induce ribozyme activity.
