Cell mass predicted by statistical analysis was 9.76 g/L after 20 h of cultivation. In a 2.5 L stirred tank reactor (STR), the cell mass produced in batch culture was 36.5 ∼39.3 g/L. Maximum cell mass in the fed-batch cultures of Saccharomyces cerevisiae JUL3 was 95.7 g/L using 50 % molasses solution and a feed rate of 10 mL/h. Cell mass obtained in fed-batch cultures was 2.4-fold that obtained in batch culture. The most efficient method of cell disruption and extraction was found to be French press and dimethyl-sulfoxide. Optimized conditions of cell disruption with French press process was 18,000 psi, 2 pass and 1min and then released protein concentration was 5.78 mg/mL. The disruption efficiency of French press was 9.17 times higher than that of other disruption methods such as ultrasonication, bead beater.
