Urushiol congener 성분들의 제거율은 영지버섯, 잔나비걸상버섯 등에 의해 각각 93%, 91%, 89%를 보였다. 옻피배지에 부영양원 (쌀) 의 첨가는 균체성장 및 urushiol의 생 전환율을 5% 상승시켰다. 2. 장수버섯이 생산하는 laccase 정제 및 특성 Urushiol의 제거는 laccase활성에 의존적이었으며 배양액 중 urushiol의 첨가는 laccase 활성과 균체량의 증가를 가져왔다. 배지성분 중 Cu^(2+)이온이 laccase 활성에 주요한 인자로 나타났다. 장수버섯은 두 종류의 laccase (Lac1, Lac2) 를 생산하였고 각각 67 kDa (Lac1), 66 kDa (Lac2) 의 분자량을 가지는 단량체 이었으며 3.67 (Lac1), 3.81 (Lac2) 의 등전점을 가지고 있었다. 정제된 laccase의 활성은 366.27 U/mg protein (Lac1), 680.34 U/mg protein (Lac2) 으로 조효소에 대하여 각각 241.0배 (Lac1) 와 447.6배 (Lac2) 정도의 정제배수를 보였다. 정제된 효소는 pH 4.5~5.0, 온도는 30~35℃에서 최대 활성을 나타내었고 두 효소 모두 Fe^(2+), Mg^(2+)에 의해서 강한 저해활성을 보였다. Na (50 mM) 이온에 의해서도 84% 이상, 0.01 mM sodium azide에 의해서 높은 저해활성을 보였다. 3. 생 전환 옻피의 생리활성 옻피 중 phenolic acid는 syringic acid와 gallic acid가 주요성분이었으며 버섯균의 증식에 의해서 79.3%/77.2% (water/70% EtOH ex.) 감소되었으나 총 아미노산 함량은 2.1배 증가하였다. 생옻피 추출물의 NIH3T3세포에 대한 세포독성 (IC_(50)) 은 약 20 μg/well (200 μL) 정도이었으나 생 전환된 옻피 추출물은 2.5배 이상 세포독성이 감소하였다. 생 전환 옻피 추출물은 NIH3T3세포에 인위적으로 유도된 oxidative stress에 대하여 세포독성이 없는 수준에서 보호효과가 있었으며 10 μg/well 농도에서 3T3-L1 전구지방세포를 가장 효과적으로 분화시켰다. 추출물을 실험동물 (rat) 에 10 mg/kg농도로 투여한 결과 뇌의 BDNF (Brain derived neurotrophic factor) 및 GDNF (glial cell line derived neurotrophic factor) 의 mRNA 및 단백질을 4.5배 (4 hr) 높게 발현시켰다. TH (tyrosine hydroxylase) mRNA 2시간 경과 후에 강하게 유도시켰으며 단백질은 5.2배 (2 hr) 높게 발현시켰을 뿐 아니라 TH-IR (immunoreactive neuron) 을 활성화시켰다. 4. 생 전환 옻피의 대량생산 및 식품에의 적용 장수버섯균 배양을 위한 액체배지로는 맥아즙배지 (GMEB) 가 다른 배지에 비해 1.7~3.8배 높은 균체생산성과 10배 정도 높은 laccase활성을 보였다. 생 전환 옻피를 대량생산하기 위한 방법으로는 옻피를 물에 하룻동안 수침한 후에 흡수되지 못한 수분을 제거하고 버섯 배양병에 담아 25℃에서 15일 동안 증식시킨 후 건조를 행하는 것이 적절하였다. 주류 제조시 생 전환 옻의 첨가는 alcohol 생성능과 색도 변화에는 영향을 미치지 않는 것으로 나타났다. 침출주 제조시 침출용 증류주의 alcohol 농도에 따른 향기성분이나 색도의 차이는 거의 보이지 않았으며 25% 침출주의 기호도가 가장 높았다. 음료 제조시에도 종래의 음료수 제품과 큰 차이가 없었다.

Rhus verniciflua Stokes (RVS) has been used in herbal medicine for diabetes mellitus and stomach disease for thousands of years in Korea in spit of containing the urushiol, an allergen in plant. Mushrooms produce a wide range of extracellular enzymes including oxidoreductase that can degrade complex organic substrates to some compounds for their nutrition. Therefore they are expected to play an important role in bioconversion/removal of potential inhibitory phenolic compounds present in the substrate. A new biological approach is tried the removal of urushiol from RVS using mushroom. 1. Biological conversion of urushiol from RVS with mushroom species All mushroom species (11 sp.) employed in this study were able to grow on RVS, although the growth rate (mm/day) was lower than control (sawdust). A thin band/spot was detected on TLC for F. fraxinea, G. lucidum and G. applanatum indicating that these species have high urushiol degrading capacity. The components of urushiol congeners [C15:3 (m/z 314), C15:2 (m/z 316), C15:1 (m/z 318), C15:0 (m/z 320)] were purified by HPLC and identified by LC-MS and GC-MS. A C15:3 (3-pentadecatrienly catechol) was found to be most abundant in RVS. F. fraxinea decreased remarkably urushiol analogues from 154.15 to 10.73 mg/100 g dry base (approximate 93%), whereas C. vercicolor showed only 1.46% of degrading capacity in spite of 2 fold higher specific growth rate. Similarly, laccase activity was found to be high in F. fraxinea but low in C. vercicolor. Moreover, approximately 98% of bioconversion was accomplished by F. fraxinea cultivated on RVS supplemented with 20% (w/w) rice bran. These findings suggest that mushrooms can be used in bioconversion of RVS. 2. Purification and characterization of laccase from F. fraxinea Urushiol content was dependent of laccase activity from F. fraxinea and it's supplementation in culture broth increased their activity and cell mass, where a Cu^(2+) major element their activity. Two protein with laccase activity were purified to homogeneity from basal medium. Both enzyme are monomeric proteins, with molecular weight around 67 kDa (Lac1) and 66 kDa (Lac2) and isoelectirc points of 3.67 and 3.81, respectively. They showed similar activity with optimum at pH (4.5~5.0) and temperature (30~35℃). The enzymes were purified approximately 241.0 (366.27 U/mg protein) and 447.6 (680.34 U/mg protein)-fold from extracellular culture, and were strongly inhibited by Fe^(2+), Mg^(2+), Na^(+) and sodium azide. 3. Biological activity of bioconverted RVS Syringic and gallic acid were predominant in RVS and acting as an antioxidant, and were decreased by the growth of F. fraxinea. But RVS cytotoxicity for NIH3T3 cell line was decreased by culturing of F. fraxinea. Antioxidant activity of bioconverted-RVS extracts against DPPH radical and NIH3T3 cell line was investigated. The extract showed protective effect on NIH3T3 cell against UV-irradiated induced oxidative stress without cytotoxicity for cell. The extract apparently enhanced the accumulation of intracellular lipid droplet at 10 μg/well (200uL), and actively inhibited cancer cell (HeLa and MCF-7) growth within 20 μg /well (200 μL). The extracts treatment induced BDNF (brain derived neurotrophic factor), GDNF (glial cell line derived neurotrophic factor) and TH (tyrosine hydroxylase) mRNA and protein expression of both in rat brain as well as activation of TH-IR (immunoreactive neuron). Our research indicates that bioconverted RVS can be developed as a therapeutic agent and might help to find the alternative therapy for the treatment of Parkinson’s disease. 4. Mass production of bioconverted RVS and their application to food F. fraxinea produced 10 times higher laccase activity as well as 1.7~3.8 times higher cell mass by culturing on GMEB (germinated malt extract broth). For mass production of bioconverted RVS, RVS was soaked in water for 1 days, and then extra water was removed from RVS and incubated at 25℃ for 15 days. Bioconverted RVS did not affect alcohol production and color of rice wine as well as volatile compound of liquor with various alcoholic concentration. This fashion similar at potable water supplemented with bioconverted RVS.
