If abnormality such as dysplasia, carcinoma, and diabetes in biological tissues are occurred, variations of refractive index mismatch between intracellular fluid and extracellular fluid is occurred. Additionally, those abnormalities increase nuclear size of cell and decrease structural protein density. These phenomena bring on scattering coefficient change. Therefore, measurement of scattering coefficient change is very important and may be used for differentiation of normal and abnormal tissues. Many groups have reported different methods about the measurement of scattering changes. Recently, other groups have reported the measurement of scattering changes using OCT. However, The method using OCT has some limitations because the intensity is relatively sensitive to the change of the focal position. Therefore, in this study, we propose a method using PS-OCT less sensitive for the focal position changes and stably able to measure scattering changes.PS-OCT is non-invasive imaging technique that allows the acquisition of high-resolution tomographic images and can provide not only cross-sectional images but also polarization states of biological tissues. In this study, we analyzed DOP acquired by PS-OCT using Boltzmann fit. In results, we found that DOP was changed by scattering and less sensitive for changes of focal position. We also found that DOP was linearly changed by changes of scattering in Intralipid suspensions and gelatine solid phantoms. Finally, we found that DOP was changed by increase of glucose concentration. This result shows that it is possible to measure glucose concentration using PS-OCT. In further study, if probe PS-OCT is made, it may be possible to measure glucose concentrations in clinical experiments.
